Neutron encoded derivatization of endothelial cell lysates for quantitation of aldehyde metabolites using nESI-LC-HRMS

Michael Armbruster et al.

Summary: Chemical derivatization with stable isotope tags is an effective method for analyzing biological aldehydes in complex samples, allowing for sample multiplexing, increased throughput, and improved signal intensity for quantitation. The use of nine quaternary amine tags and low-concentration perfluoropentanoic acid as an ion pairing reagent resulted in accurate analysis of multiple aldehydes with improved analyte retention and detection limits.

Chloramine-T-Enabled Mass Spectrometric Analysis of C=C Isomers of Unsaturated Fatty Acids and Phosphatidylcholines in Human Thyroids

Bo Zhang et al.

Summary: Specific locations of carbon-carbon double bonds (C=C) in lipids play important roles in biological processes. A N-tosylaziridination method was developed to annotate C=C positions in lipids using mass spectrometry, and it showed high efficiency, sensitivity, and specificity. Studies on human thyroid tissues suggested that increased unsaturated fatty acid content and n-6/n-3 ratios were closely associated with thyroid tumorigenesis.

Simultaneous multiplexed quantification and C=C localization of fatty acids with LC-MS/MS using i(s)under-barobaric m(u)under-barltiplex rea(g)under-barents for c(a)under-barrbonyl-containing compound (SUGAR) tags and C=C epoxidation

Ting-Jia Gu et al.

Summary: This study developed a high-throughput analysis method for simultaneous quantification and determination of the positions of carbon-carbon double bonds (C=C) in unsaturated fatty acids (FAs). The method involved epoxidation and labeling, combined with liquid chromatography-mass spectrometry (LC-MS), to achieve quantitative and positional analysis of different unsaturated FA isomers. The method demonstrated excellent performance in complex sample analysis.

Enhancing the Signal-to-Noise of Diagnostic Fragment Ions of Unsaturated Glycerophospholipids via Precursor Exclusion Ultraviolet Photodissociation Mass Spectrometry (PEx-UVPD-MS)

Samuel W. J. Shields et al.

Summary: Understanding the structures and functions of lipids is important in lipidomics research. This study introduces a technique called precursor exclusion (PEx) UVPD, which improves the signal-to-noise ratio of low-abundance fragment ions obtained from UVPD of glycerophospholipids. The method allows for deeper structural analysis of lipids and confident identification of different lipid structures.

Imaging and Analysis of Isomeric Unsaturated Lipids through Online Photochemical Derivatization of Carbon-Carbon Double Bonds

Daisy Unsihuay et al.

Summary: The development of a novel online photochemical approach for lipid isomer identification utilizing selective derivatization of double bonds with singlet oxygen provides a new strategy for exploring the complexity of the lipidome. Fragmentation of lipid hydroperoxide products in a mass spectrometer allows for rapid identification of C=C bond locations in lipids. This cost-effective method can be easily integrated into lipidomics workflows.

Summary: This study developed a workflow for global quantitation of fatty acids, including C=C location isomers, via charge-tagging Patern`o-Buchi (PB) derivatization and liquid chromatography-tandem mass spectrometry (LC-MS/MS). The method enabled deep profiling of unsaturated fatty acids in yak milk powder and pooled human plasma, achieving successful quantitation across a wide range of concentrations.
