Rapid shear stress-dependent ENaC membrane insertion is mediated by the endothelial glycocalyx and the mineralocorticoid receptor

Summary: The contribution of shear stress-sensitive ENaC to the mechanical properties of endothelial cell surface in (patho)physiological conditions was investigated. It was found that acute laminar shear stress led to the insertion of ENaC and subsequent stiffening of the endothelial cortex. Non-laminar shear stress further increased the stiffness of the cells. These effects depended on the presence of the endothelial glycocalyx and could be prevented by inhibiting ENaC and MR. In C57BL/6 vessels, inhibiting ENaC and MR blunted flow- and acetylcholine-mediated vasodilation, while in ApoE/LDLR-/- vessels, this effect was absent. In conclusion, ENaC and the glycocalyx were identified as important shear stress sensors and mediators of endothelium-dependent vasodilation under physiological conditions, whereas their role was lost in pathophysiological conditions, contributing to endothelial stiffening and dysfunction.

Uptake of oxidative stress-mediated extracellular vesicles by vascular endothelial cells under low magnitude shear stress

Summary: This study demonstrates that low magnitude shear stress promotes the uptake of red blood cell-derived extracellular vesicles (RBCEVs) by endothelial cells in vitro and in animal models. Oxidative stress is identified as a mechanism for the increased uptake of RBCEVs. These findings provide a theoretical basis for the development and utilization of EVs-based nano-drug delivery systems in vivo.

Extracellular matrix regulates force transduction at VE-cadherin junctions

Summary: Increased tension on VE-cadherin complexes activates adaptive cell stiffening and local cytoskeletal reinforcement, which are key features of intercellular mechano-transduction. This study shows that pulling on VE-cadherin receptors initiates a cascade that activates integrins and strengthens stressed cadherin adhesions. This cascade is different from the antagonistic effects of integrins on intercellular junctions. The involvement of integrins sensitizes intercellular force transduction to the extracellular matrix, altering signal cascades that reinforce cell-cell adhesions.
