The Role of Decorin Proteoglycan in Mitophagy

Thomas Neill et al.

Summary: This review explores the role of decorin, a small leucine-rich proteoglycan, in autophagy and mitochondrial autophagy in breast cancer. Decorin interacts with cell surface receptor tyrosine kinases, VEGFR2 and MET receptor, to induce autophagy and mitophagy, respectively. Understanding the mechanisms underlying intracellular catabolism regulation provides new insights for protein therapies targeting autophagic progression.

In silico and in vitro mapping of specificity patterns of glycosaminoglycans towards cysteine cathepsins B, L, K, S and V

K. K. Bojarski et al.

Summary: This study investigated the interactions between human cysteine cathepsins and various types of glycosaminoglycans (GAGs). Molecular docking simulations and molecular dynamics analysis revealed that electrostatics play a crucial role in driving the cathepsin-GAG interactions, and the length of GAG chains significantly affects the strength of these interactions. Furthermore, binding of GAGs at different regions on cathepsins surface has varying effects on the enzymatic activity, depending on the cathepsin and GAG type.

Biglycan Interacts with Type I Insulin-like Receptor (IGF-IR) Signaling Pathway to Regulate Osteosarcoma Cell Growth and Response to Chemotherapy

Eirini-Maria Giatagana et al.

Summary: Osteosarcoma is an aggressive bone cancer, and biglycan plays a role in promoting its growth and resistance to chemotherapy by activating the IGF-IR signaling pathway. This mechanism varies in different levels of differentiation in osteosarcoma cells.

UniProt: the universal protein knowledgebase in 2021

Alex Bateman et al.

Summary: The UniProt Knowledgebase aims to provide users with a comprehensive, high-quality set of protein sequences annotated with functional information. Updates over the past two years have increased the number of sequences to approximately 190 million, with new methods to assess proteome completeness and quality. UniProtKB has responded to the COVID-19 pandemic by expertly curating relevant entries and making them rapidly available through a dedicated portal.

The Role of Decorin and Biglycan Signaling in Tumorigenesis

Valentina Diehl et al.

Summary: Decorin and biglycan play critical roles in tumor biology by influencing key processes that affect tumor growth and progression. Despite their structural homology, these proteoglycans interact with different cell surface receptors, modulating distinct signaling pathways that ultimately impact cancer development.

The Extracellular Small Leucine-Rich Proteoglycan Biglycan Is a Key Player in Gastric Cancer Aggressiveness

Filipe Pinto et al.

Summary: Approximately 80% of gastric cancer patients are diagnosed at advanced stages with poor prognosis. The aggressiveness of gastric cancer is associated with biglycan, an extracellular proteoglycan, which requires further research to explore its therapeutic potential.

Matrix lumican endocytosed by immune cells controls receptor ligand trafficking to promote TLR4 and restrict TLR9 in sepsis

George Maiti et al.

Summary: The ECM protein lumican plays a significant role in regulating infections and inflammation, promoting TLR4 response while restricting TLR9 in immune cells. Lumican may be internalized by immune cells to modulate their transcriptional programs and effector responses, suggesting potential therapeutic applications in future treatments.
