Engineered strategies to enhance tumor penetration of drug-loaded nanoparticles

Mohammad Souri et al.

Summary: Nanocarriers have been widely studied for anticancer drug delivery, with their clinical translation facing challenges due to inefficient accumulation and penetration in tumor microenvironment. Despite various strategies proposed to overcome these challenges, their success has been limited so far. Novel strategies such as transformable nanocarriers and transcellular transport of peptide-modified nanocarriers have emerged as potential advanced drug delivery systems.

Transcytosis-enabled active extravasation of tumor nanomedicine

Quan Zhou et al.

Summary: Extravasation is the crucial process for nanomedicines to reach solid tumors. However, the Enhanced Permeability and Retention (EPR) effect, which was previously considered as the primary extravasation mechanism, is limited and heterogeneous in human tumors. To overcome this limitation and enhance therapeutic efficacy, the development of transcytosis-inducing nanomedicines has been proposed. These nanomedicines can actively extravasate into solid tumors by utilizing the transcellular transport mechanism of tumor endothelial cells.

Effect of Cationic Charge Density on Transcytosis of Polyethylenimine

Siqin Chen et al.

Summary: This study investigates the effect of cationic charge on adsorption-mediated transcytosis-induced tumor penetration, finding that PEI with 24% acetyilation has the highest transcytosis efficiency due to its balanced cell-binding affinity. The findings highlight the critical influence of surface cationic charge density on tumor penetration efficiency, providing insights for designing drug delivery systems with active transcytosis for enhanced therapeutic effects.
