3D-Printed PCL/Zn scaffolds for bone regeneration with a dose-dependent effect on osteogenesis and osteoclastogenesis

Summary: In this study, PCL/Zn composite scaffolds were successfully fabricated by adding different Zn powder contents, which significantly improved the mechanical properties and cytocompatibility of the scaffolds. In vivo experiments demonstrated that the addition of Zn powder promoted new bone formation in a dose-dependent manner. Additionally, Zn2+ was found to regulate osteogenesis and osteoclastogenesis through the activation of different signaling pathways.

Results showed the scaffolds significantly enhanced bone mesenchymal stem cells' osteogenic differentiation and chondrocytes' proliferation, leading to improved cartilage and subchondral bone repair in rabbit osteochondral defects after 8- and 12-week implantation. The findings suggest that the insulin-PLGA/PDA/PCL scaffolds may be a promising strategy for osteochondral repair.
