Recent advances in human respiratory epithelium models for drug discovery

Naheem Yaqub et al.

Summary: The respiratory epithelium is important in viral infections and chronic illnesses. Current model systems to study respiratory diseases have limited correlation to the human condition, resulting in low success rates for drug development. New respiratory models are needed to address this problem.

Enhanced drug delivery to the reproductive tract using nanomedicine reveals therapeutic options for prevention of preterm birth

Hannah C. Zierden et al.

Summary: Inflammation contributes to nearly 4 million global premature births annually, and current clinically used treatments with progesterone are not effective in preventing inflammation-induced preterm birth. However, vaginal delivery of mucoinert nanosuspensions of histone deacetylase (HDAC) inhibitors, in some cases with the addition of progesterone, can prevent preterm birth in a mouse model and result in the birth of live pups exhibiting neurotypical development.

Mucoadhesive hyaluronic acid-based films for vaginal delivery of metronidazole

Marina L. Gerton et al.

Summary: The newly developed hyaluronic acid film for delivering metronidazole offers a more effective and convenient alternative treatment for bacterial vaginosis, with the ability to gradually release the drug in the vaginal environment for an extended period. This innovative approach addresses the shortcomings of current treatments, providing a promising solution for women's health.

Organoids of the female reproductive tract

Cindrilla Chumduri et al.

Summary: The healthy functioning of the female reproductive tract relies on balanced hormonal regulation during different reproductive stages. The mucosal epithelial lining in various regions of the FRT enables selective transport of gametes and successful transfer of the zygote. Three-dimensional organoid systems from the FRT now offer valuable experimental models for studying normal physiology and disease.

Investigation of infertility using endometrial organoids

Konstantina Nikolakopoulou et al.

Summary: Infertility is a common issue in modern societies, with therapeutic interventions such as medical treatment, surgical procedures, or ART often needed. Understanding the dynamic changes of the endometrium in preparing for pregnancy is challenging, with defects in the processes of endometrial repair, decidualization, and acquisition of receptivity potentially contributing to infertility. Organoid models offer a promising approach to studying the cellular and molecular mechanisms underlying non-pregnant endometrium and early pregnancy, potentially leading to transformative therapeutic applications for reproductive failure.

Bacterial vaginosis and health-associated bacteria modulate the immunometabolic landscape in 3D model of human cervix

Pawel Laniewski et al.

Summary: This study demonstrated that 3D cervical epithelial cells colonized with cervicovaginal microbiota faithfully reproduce the immunometabolic microenvironment previously observed in clinical studies and can successfully be used as a robust tool to evaluate host responses to commensal and pathogenic bacteria in the female reproductive tract.

Summary: Current treatment options for diseases and dysfunctions of the female reproductive tract tissues are limited, necessitating exploration of alternative therapeutic approaches. Regenerative medicine technologies, such as the use of stem cells, biomaterial scaffolds, and bio-printing, may provide new avenues for therapy, which could be used to repair functional issues in female reproductive tissues.

Next generation strategies for preventing preterm birth

Hannah C. Zierden et al.

Summary: Preterm birth is defined as delivery before 37 weeks of gestation, with 15 million infants born prematurely globally each year, increasing the risk of mortality and health challenges. While the only FDA approved therapy for preventing preterm birth in the U.S. has faced recent setbacks, interdisciplinary efforts are underway to develop next generation therapies to prevent preterm birth and save infant lives. This research aims to better identify women at risk for preterm birth, prevent premature labor, and ultimately improve outcomes for newborns.

Testing of aerosolized ciprofloxacin nanocarriers on cystic fibrosis airway cells infected with P. aeruginosa biofilms

Jenny Juntke et al.

Summary: The major pathogen in adult cystic fibrosis patients is Pseudomonas aeruginosa, which forms antibiotic-resistant biofilms. A new in vitro model with human bronchial epithelial cells infected with P. aeruginosa biofilm at the air-liquid interface (ALI) was developed to test aerosolized anti-infective nanocarriers. Aerosolized ciprofloxacin-loaded nanocarriers applied 1 hour post-infection eradicated all planktonic bacteria and significantly reduced the biofilm fraction of the pathogen.
