MMV020275 and MMV020490, promising compounds from malaria box for the treatment of equine piroplasmosis

Summary: This study evaluated the inhibitory effects of MMV Malaria Box compounds on equine piroplasmosis and discovered new potent antipiroplasm drugs, including MMV020275 and MMV020490. MMV020275 significantly down-regulated the mRNA levels of the PKG gene. Clofazimine enhanced the inhibitory efficacy of MMV compounds.

Evaluation of the inhibitory effect of Zingiber officinale rhizome on Babesia and Theileria parasites

Summary: The study showed that the methanolic extract of Zingiber officinale rhizome (ZOR) has inhibitory effects on the in vitro growth of bovine Babesia and equine piroplasm parasites, with B. divergens being the most susceptible. Additionally, the combination of ZOR with diminazene aceturate (DA) or anti-B. bigemina MMV hits enhanced the inhibitory activity against these parasites, making ZOR a potential medicinal plant for babesiosis treatment.

Impact of using pyronaridine tetraphosphate- based combination therapy in the treatment of babesiosis caused by Babesia bovis, B. caballi, and B. gibsoni in vitro and B. microti in mice

Summary: The study evaluated the inhibitory effects of pyronaridine tetraphosphate (PYR) in combination with clofazimine (CF) and MMV396693 against various Babesia parasites. Results showed that PYR combined with MMV396693 had additive and indifferent interactions against Babesia parasites, while PYR combined with CF had indifferent and antagonistic interactions against certain Babesia species. Combination therapy of PYR-CF showed significant inhibition of parasite growth in mice compared to control. Treatment with PYR-CF also exhibited greater inhibition of B. microti compared to PYR alone.
