Summary: Pyroptosis is a highly inflammatory and lytic type of programmed cell death initiated by inflammasomes, with recent studies linking a family of pore-forming proteins called gasdermins (GSDMs) to this process. GSDMs have characteristic features of forming pores in the plasma membrane by releasing N-terminal fragments that can bind to acidic lipids, leading to disruption of cellular homeostasis and cell death induction. Recent studies have also revealed the structural and molecular aspects of autoinhibition, as well as the functional consequences of gasdermin-induced membrane permeabilization.

Pyroptosis executive protein GSDMD as a biomarker for diagnosis and identification of Alzheimer's disease

Summary: The levels of GSDMD, T-Tau, and Tau181p were higher in AD patients compared to VD patients and healthy controls, while A beta(1-42) levels were lower in AD patients. GSDMD showed good diagnostic accuracy in AD and good discrimination accuracy between AD and VD. The expression levels of inflammatory factors (IL-1 beta and IL-6) in the cerebrospinal fluid were higher in AD patients and positively correlated with GSDMD expression.
