Inductive Remodeling of Extracellular Matrix Scaffolds in the Temporomandibular Joint of Pigs

Bryan N. Brown et al.

Summary: This study investigates the use of an acellular scaffold composed of extracellular matrix (ECM) derived from small intestinal submucosa (SIS) as a regenerative template for the TMJ disc in a porcine model. The results show that the acellular bioscaffold filled a medical need for which there is currently no effective treatment and may represent a clinically relevant off-the-shelf implant for reconstruction of the TMJ disc.

Summary: The lack of vascularization in the white-red and white zone of the meniscus hinders its self-healing capacity and may accelerate osteoarthritis. This study developed a hybrid construct as a meniscus tissue engineering scaffold with good regenerative potential. The scaffold showed promising results in both in vitro and in vivo experiments.

Decellularized extracellular matrix scaffolds: Recent trends and emerging strategies in tissue engineering

Xuewei Zhang et al.

Summary: The application of decellularized extracellular matrix (dECM) scaffolds holds enormous potential for tissue regeneration. These scaffolds provide an optimal non-immune environment with native three-dimensional structures and bioactive components, making them an ideal choice for regenerating functional organs/tissues, especially when combined with stem cells.

The resurgent landscape of xenotransplantation of pig organs in nonhuman primates

Xuan Zhang et al.

Summary: Organ shortage in allotransplantation is a major issue leading to patient deaths or transplant ineligibility. Genetically engineered pig organs have shown some progress as potential alternatives, but still face challenges such as rejection and inflammation. Further genetic modifications and pretransplant screenings are necessary for successful xenotransplantation.

Overloading stress-induced progressive degeneration and self-repair in condylar cartilage

Lingli Fang et al.

Summary: Overloading stress induces progressive degeneration in TMJ cartilage, but can be reversed by self-repair at early stages. During the initial adaptation stage, robust proliferation of cartilage occurs, followed by widespread chondrocyte apoptosis and subsequent new chondrocyte proliferation with matrix degradation and subchondral bone catabolism.

Risk Factors Associated With Poor Outcomes Following Temporomandibular Joint Discectomy and Fat Graft

Owen G. Ellis et al.

Summary: This study aimed to estimate the incidence and risk factors associated with poor postoperative outcomes following temporomandibular joint (TMJ) discectomy and abdominal fat grafting. The study included 129 patients who underwent 132 TMJ discectomies, with a success rate of 75.2% and a conversion rate to TMJ total joint replacement (TJR) of 11.7%. Results suggest that lower improvement in mouth opening at 1 year following surgery is likely to result in failure of the TMJ discectomy procedure.

Mechanical characterization and viscoelastic model of the ovine temporomandibular joint Disc in indentation, uniaxial tension, and biaxial tension

Kevin M. Labus et al.

Summary: Recent investigations have focused on developing disc replacements and regenerative medicine for the internal derangements of the temporomandibular joint (TMJ) disc. This study characterized the mechanical behavior of the ovine TMJ disc, finding significantly higher stiffness in tension in the anteroposterior direction and a hyper-viscoelastic constitutive model capturing dynamic stress-stretch behavior. These findings support computational modeling of TMJ mechanics, biomaterials identification, and the development of implant designs for TMJ disc replacement.
