Application of extracellular vesicles derived from mesenchymal stem cells as potential therapeutic tools in autoimmune and rheumatic diseases

Huldani Huldani et al.

Summary: Mesenchymal stem cells (MSCs) have great potential as therapeutic candidates, but their clinical application is limited due to their tumorigenic properties. Extracellular vesicles (EVs), as paracrine factors of MSCs, have gained attention as potential cell-free replacement therapy due to their low immunogenicity and tumorigenicity, as well as easy management.

Anti-sclerostin antibodies and abaloparatide have additive effects when used as a countermeasure against disuse osteopenia in female rats

Mikkel Bo Brent et al.

Summary: This study compared the effectiveness of romosozumab and abaloparatide in preventing bone loss induced by botulinum toxin. The results showed that the combination of Scl-Ab and ABL maximized osteoanabolic effects and counteracted the negative skeletal effects of disuse.

Advances in pathogenesis and therapeutic strategies for osteoporosis

Shasha Song et al.

Summary: Osteoporosis, the most common bone disorder globally, is characterized by low bone mineral density and increased fracture risk. The pathogenesis of osteoporosis involves various factors, including the gut microbiome, autophagy, iron balance, and cellular senescence. Promising drugs targeting these mechanisms, such as probiotics, alpha-ketoglutarate, senolytics, and hydrogen sulfide, have been identified for osteoporosis treatment.

A Systematic Review of Animal Models of Disuse-Induced Bone Loss

Mikkel Bo Brent et al.

Summary: Multiple animal models of disuse-induced bone loss exist, with rats (59%) and mice (30%) being the most commonly used species. Hindlimb unloading (44%), neurectomy (15%), bandages and orthoses (15%), and botulinum toxin (9%) were the most frequently used methods to induce disuse. The median time of disuse was 21 days (quartiles: 14 days, 36 days), with a median number of animals per group subjected to disuse being 10 (quartiles: 7, 14).

Advances in mesenchymal stem cell transplantation for the treatment of osteoporosis

Yuhe Jiang et al.

Summary: Osteoporosis is a systemic metabolic bone disease characterized by bone loss and microstructural degeneration, leading to increasing personal and societal costs. MSC transplantation shows promising potential in treating osteoporosis, but issues such as safety, transplant efficiency, and standardization of the manufacturing process need to be addressed. Comprehensive evaluation of clinical trials is essential for clinical translation of MSC therapy for osteoporosis.

Muscle-derived exosomes encapsulate myomiRs and are involved in local skeletal muscle tissue communication

Chrystalla Mytidou et al.

Summary: Exosomes are extracellular vesicles released from cells, mediating cell-to-cell and tissue-to-tissue communication in normal and pathological conditions. Muscle tissues secrete exosomes containing specific miRNAs, which vary in sorting between skeletal muscles of different fiber types, influencing local and systemic communication between muscles.

Protein profile of fiber types in human skeletal muscle: a single-fiber proteomics study

Marta Murgia et al.

Summary: This study utilized single-fiber proteomics to identify 404 proteins with significant differences among different fiber types, including many specific markers for type 1 or 2X fibers. It also revealed other major patterns of protein distribution and highlighted the selective enrichment of certain proteins in specific fiber types, suggesting potential implications in muscle pathophysiology. Single-fiber proteomics, combined with emerging single-cell proteomics approaches, are crucial for exploring and quantifying muscle cell heterogeneity.
