Combinatorial Approach of Binary Colloidal Crystals and CRISPR Activation to Improve Induced Pluripotent Stem Cell Differentiation into Neurons

Summary: Conventional methods of neuronal differentiation in human induced pluripotent stem cells are complicated, but using artificial extracellular matrices can improve in vitro neuronal differentiation. This study tested the impact of binary colloidal crystals on neuronal differentiation and found that fine tuning particle size can improve neuronal differentiation and branching. Binary colloidal crystals with 5μm silica and 100nm carboxylated polystyrene had the most prominent effect on increasing neurite outgrowth and more complex ramification.

Programmed Topographic Substrates for Studying Roughness Gradient-Dependent Cell Migration Using Two-Photon Polymerization

Summary: This study explores the sensing and modulation of cell migration velocity and direction in response to spatial roughness. Three characteristic cell migration behaviors were identified: higher migration velocity on substrates with higher roughness, preference for migration from regions of higher roughness to lower roughness, and unchanged migration velocity on a lower roughness range on a graded substrate.

Biophysical Regulations of Epigenetic State and Notch Signaling in Neural Development Using Microgroove Substrates

Summary: Surface topography, such as microgrooves, can alter the behavior and differentiation patterns of stem cells by enhancing alignment, modifying nuclear geometry, increasing cellular stiffness, and inducing epigenetic changes. In this study, microgrooves were found to promote neuronal differentiation of neural stem cells by downregulating Notch signaling and modifying the epigenetic landscape.
