Design and outcome measures of LAVENDER, a phase 3 study of trofinetide for Rett syndrome

Jeffrey L. Neul et al.

Summary: A drug trial using trofinetide in females with Rett syndrome (RTT) showed significant improvement compared to placebo on multiple measures. A Phase 3 trial will further evaluate the efficacy and safety of trofinetide using disease-specific and novel scales to assess core symptoms of RTT, quality of life, and caregiver burden.

Glycyl-L-Prolyl-L-Glutamate Pseudotripeptides for Treatment of Alzheimer's Disease

Hasan Turkez et al.

Summary: This study investigated the neuroprotective potentials of three novel GPE peptidomimetics in an in vitro AD model, showing that these analogs modulate oxidative stress, ACh depletion, alpha-secretase inactivation, apoptotic, and necrotic cell death. Therefore, the treatments with these novel GPE analogs might be promising therapeutic agents for treatment and/or prevention of AD.

Macrocyclization strategies for cyclic peptides and peptidomimetics

Clement Bechtler et al.

Summary: Peptides are increasingly used as a therapeutic class due to their unique spatial characteristics which can target difficult-to-drug protein-protein interactions and surfaces. Despite their advantages, peptides face challenges such as high conformational flexibility and susceptibility to proteolytic cleavage. Macrocycle formation from linear peptides can help overcome these challenges and improve their characteristics.
