Paracrine FGFs target skeletal muscle to exert potent anti-hyperglycemic effects

Lei Ying et al.

Summary: This study identifies paracrine FGF4 as an anti-hyperglycemic FGF that targets skeletal muscle to increase the abundance of the glucose transporter GLUT4 on the cell surface. Compared to endocrine FGF21, the paracrine FGF4 shows superior glycemic control in diabetic mice and has anti-inflammatory properties, suggesting its potential for treating type 2 diabetes and related metabolic disorders.

aFGF alleviates diabetic endothelial dysfunction by decreasing oxidative stress via Wnt/β-catenin-mediated upregulation of HXK2

Jia Sun et al.

Summary: The study showed that aFGF alleviates diabetic endothelial dysfunction by suppressing mitochondrial oxidative stress through the Wnt/beta-catenin/c-Myc axis and HXK2. The specific mechanisms by which aFGF regulates these processes remain unclear.

Highly accurate protein structure prediction with AlphaFold

John Jumper et al.

Summary: Proteins are essential for life, and accurate prediction of their structures is a crucial research problem. Current experimental methods are time-consuming, highlighting the need for accurate computational approaches to address the gap in structural coverage. Despite recent progress, existing methods fall short of atomic accuracy in protein structure prediction.

Highly accurate protein structure prediction for the human proteome

Kathryn Tunyasuvunakool et al.

Summary: Using the AlphaFold method, the structural coverage of the proteome has been significantly expanded, covering 98.5% of human proteins with 58% of residues having confident predictions and 36% having very high confidence. Introducing new metrics to interpret the dataset and identify disordered regions, this study aims to provide high-quality predictions for generating biological hypotheses.

High-accuracy protein structure prediction in CASP14

Joana Pereira et al.

Summary: The application of state-of-the-art deep-learning approaches to protein modeling problem has expanded the high-accuracy category in CASP14, evaluating the performance of different groups and introducing new metrics. Despite the significant progress made by AlphaFold2, the second-best method in CASP14 outperformed the best method in CASP13, demonstrating the role of community-based benchmarking in the development of protein structure prediction field.

FGF1ΔHBS prevents diabetic cardiomyopathy by maintaining mitochondrial homeostasis and reducing oxidative stress via AMPK/Nur77 suppression

Dezhong Wang et al.

Summary: Research has shown that FGF1 levels are decreased in DCM patients and positively correlated with fraction shortening, potentially making it a therapeutic target. Treatment with FGF1 variant FGF1( increment HBS) can prevent diabetes-induced cardiac injury and remodeling through effects on anti-oxidative gene expression and Nur77 levels. FGF1( increment HBS) exerts its beneficial effects by reducing mitochondrial fragmentation, ROS generation, and cytochrome c leakage, as well as enhancing mitochondrial respiration and beta-oxidation, all in an AMPK/Nur77-dependent manner.
