Tunable Surface Charge Enables the Electrostatic Adsorption- Controlled Release of Neuroprotective Peptides from a Hydrogel-Nanoparticle Drug Delivery System

Eric Ho et al.

Summary: We controlled the release of neuroprotective peptide PACAP from PLGA nanoparticles dispersed in a HAMC hydrogel by exploiting the electrostatic interactions between PACAP and nanoparticles. The release rate of PACAP was tuned by modifying the nanoparticle loading or surface charge, which was controlled by the pH responsive carboxylic acid groups of PLGA. This system successfully delivered PACAP to the central nervous system for the treatment of neurological disorders.

An injectable PEG hydrogel controlling neurotrophin-3 release by affinity peptides

Jing Wang et al.

Summary: A study was conducted to develop an injectable hydrogel delivery system for localized release of neurotrophin-3, which provides affinity-controlled release and longer delivery time compared to Poloxamer hydrogels. The injectable PEG hydrogels induced local immune responses similar to biocompatible Poloxamer hydrogels, but released payloads at a slower rate. The sustained release of neurotrophin-3 from peptide-conjugated PEG hydrogels was verified, demonstrating a reversible interaction between peptides and proteins.

Engineered bridge protein with dual affinity for bone morphogenetic protein-2 and collagen enhances bone regeneration for spinal fusion

Priscilla S. Briquez et al.

Summary: A new translational protein engineering-based approach has been developed to reduce the required dosage of rhBMP-2 in clinical spinal fusion, increasing safety when delivered in a collagen sponge. This method has shown improved efficacy at low doses of rhBMP-2 in two mouse models of bone regeneration and may have applications in various regenerative medicine fields.

Metabolic and immunomodulatory control of type 1 diabetes via orally delivered bile-acid-polymer nanocarriers of insulin or rapamycin

Jung Seok Lee et al.

Summary: Orally delivered nanocarriers made from polymeric bile acid, carrying insulin or rapamycin, demonstrate metabolic and immunomodulatory functions in restoring blood-glucose levels, delaying the onset of diabetes, reversing inflammation, restoring metabolic functions, and extending animal survival. These ingestible bile-acid-polymer nanocarriers may offer translational opportunities for the prevention and treatment of type 1 diabetes.
