Multidimensional Isotropic Magnetic Shielding Contour Maps for the Visualization of Aromaticity in ortho-Arynes and Their Reactions

Albert Artigas et al.

Summary: Visualization of electron delocalization and aromaticity in selected aryne compounds, including nonplanar examples, and their reactions was achieved using multidimensional isotropic magnetic shielding contour maps. The maps showed that aryne compounds are generally less aromatic than the corresponding arene compounds, with aromaticity peaking during the reaction when approaching the transition state.

Determination of the Rate Constant of the [4+2] Cycloaddition Between an Aryne Atropisomer and Furan in Solution

Guillaume Dauvergne et al.

Summary: By combining theoretical calculations and experimental measurements, the kinetic constant of the cycloaddition reaction between a specially designed nonracemic aryne atropisomer and furan in solution was determined. The reaction half-life of this aryne atropisomer with 100 equivalents of furan as the trapping reagent was found to be <150 ns at temperatures above -20 degrees C.

Carbo[n]helicenes Restricted to Enantiomerize: An Insight into the Design Process of Configurationally Stable Functional Chiral PAHs

Prince Ravat

Summary: The interconversion of enantiomers is a key stereodynamic feature of carbo[n]helicenes, with the Gibbs activation energy determining their configurational stability. This minireview provides an overview of the mechanism, recent developments, and factors affecting the enantiomerization of [n]helicenes, aiming to accelerate the design process of configurationally stable functional chiral molecules based on helicene substructures. Additionally, misconceptions and irregularities in the recent literature on terms such as racemization and enantiomerization, as well as the calculation of activation parameters for [n]helicenes and related compounds, are addressed.

o-Silylaryl Triflates: A Journey of Kobayashi Aryne Precursors

Jiarong Shi et al.

Summary: Arynes are highly active organic intermediates used in the rapid synthesis of substituted arenes, with o-silylaryl triflates being the most frequently employed precursors. The resurgence in aryne chemistry over the past 20 years is largely attributed to the widespread use of Kobayashi's method, which involves fluoride-induced removal of the TMS group with concomitant departure of its ortho OTf group. This review provides an overview of the history, achievements, and applications of Kobayashi's method in the synthesis of natural products, bioactive molecules, and polycyclic aromatic hydrocarbons.

Recent Advances in Catalytic Asymmetric Construction of Atropisomers

Jun Kee Cheng et al.

Summary: Atropisomerism is a stereochemical behavior exhibited by three-dimensional molecules with rotationally restricted sigma bonds, and compounds with atropisomerically chiral properties are increasingly utilized in fields where molecular asymmetry is influential. There is a steady demand for atroposelective synthesis, leading to conceptually novel and streamlined methods for expanding the structural diversity of atropisomers. This review summarizes key achievements in the stereoselective preparation of biaryl, heterobiaryl, and nonbiaryl atropisomers between 2015 and 2020, emphasizing synthetic strategies for each structural class and potential applications of atropochiral targets.

Kobayashi Benzynes as Hexadehydro-Diels-Alder Diynophiles

Sahil Arora et al.

Summary: A cascade strategy for naphthyne formation using Kobayashi benzynes in an in situ hexadehydro-Diels-Alder reaction was reported, allowing for rapid construction of various naphthalenic products. Density functional theory computations suggest that the strained benzynes act as superdiynophiles in this transformation. The trapping efficiencies of several arynophiles were determined using benzyne to naphthyne cyclization as an internal clock reaction.

Could London Dispersion Force Control Regioselective (2+2) Cyclodimerizations of Benzynes? YES: Application to the Synthesis of Helical Biphenylenes

Takashi Ikawa et al.

Summary: In recent years, it has been discovered that London dispersion interactions play an important role in controlling the regio- and stereo-selectivity of reactions. These interactions are dominant in selective catalysis, while repulsive steric interactions are predominant in noncatalytic reactions. A novel method utilizing London dispersion interactions has been developed for the synthesis of proximal biphenylenes, which have potential applications in ligands, catalysts, and medicines.

Atroposelective transformation of axially chiral (hetero)biaryls. From desymmetrization to modern resolution strategies

Jose A. Carmona et al.

Summary: This tutorial review provides a comprehensive overview of methodologies available for the atroposelective transformation of (heterobiaryl)biaryl precursors towards enantiomerically enriched products and discusses the conceptual aspects associated with each type of transformation. Several strategies have been developed based on the presence or absence of symmetry in the starting material and the occurrence of racemization or dynamicization events, including desymmetrization, classical kinetic resolution (KR), dynamic kinetic resolution (DKR), and dynamic kinetic asymmetric transformation (DYKAT). Seminal contributions and selected examples are discussed to highlight the potential of these synthetic tools.
