Dynamic Adjust of Non-Radiative and Radiative Attenuation of AIE Molecules Reinforces NIR-II Imaging Mediated Photothermal Therapy and Immunotherapy

Zhenjie Wang et al.

Summary: Due to limitations in aggregation-caused quenching effect and poor penetration capabilities in common fluorescent molecules, their applications in visualized imaging and photoactivated treatment are restricted. To overcome this, a new near-infrared II (NIR-II) molecule named TST, which possesses aggregation-induced emission (AIE) and photothermal therapy abilities, is synthesized. The molecule is co-assembled with camptothecin prodrug (CPT-S-PEG) and immune checkpoint inhibitor AZD4635 into nanoparticles for drug delivery, aiming to enhance fluorescent yield and therapeutic effect.

Summary: A versatile theranostic agent, TPA-BT-DPTQ, was designed and prepared for simultaneous fluorescence imaging, photoacoustic imaging, photothermal imaging, and thermal eradication of tumors. The photothermal therapy mediated by TPA-BT-DPTQ nanoparticles not only destroys the primary tumor but also enhances immunogenicity for further suppressing the growth of distant tumors. Combining photothermal therapy with a PD-L1 antibody prevents cancer metastasis and recurrence.

Xiaoqing Yi et al.

Summary: The study introduced a self-guiding polymeric micelle for enhanced chemo-photodynamic therapy by utilizing a dual-stage light irradiation strategy, combining two AIE photosensitizers and a reduction-sensitive paclitaxel prodrug. The first light irradiation facilitated cellular uptake, while the second light irradiation led to efficient ROS production and monitored drug release. This dual-stage light irradiation strategy showed superior therapeutic effects compared to continuous light irradiation for only 24 minutes in both in vivo and in vitro studies.
