Cryo-EM structures of prion protein filaments from Gerstmann-Straussler-Scheinker disease

Grace Hallinan et al.

Summary: The aggregation of Prion protein (PrP) and formation of PrP amyloid (APrP) are crucial events in the development of prion diseases. Gerstmann-Straussler-Scheinker (GSS) disease, a dominantly inherited form of prion protein amyloidosis, is characterized by the presence of PrP amyloid plaques throughout the brain. Cryo-EM analysis of APrP filaments isolated from GSS patients with the F198S mutation revealed their complex structure, highlighting the importance of understanding filament structures in human neurodegenerative diseases.

Structural biology of ex vivo mammalian prions

Efrosini Artikis et al.

Summary: The structures of infectious prion fibrils have been revealed using cryo-EM, providing insights into the structural features that contribute to pathogenicity. Variations in the conformation and sequence of PrP fibrils likely contribute to strain-dependent effects, while posttranslational modifications may play a role in membrane binding and pathogenesis.

Cryo-EM structure of anchorless RML prion reveals variations in shared motifs between distinct strains

Forrest Hoyt et al.

Summary: This study presents a high-resolution cryo-electron microscopy-based structure of infectious brain-derived fibrils of the mouse anchorless RML scrapie strain. Comparisons with the previously determined hamster 263K strain reveal both common and divergent features among these prion strains.

Novel quaternary structures of the human prion protein globular domain

Leandro Oliveira Bortot et al.

Summary: Prion disease is caused by the misfolding of cellular prion protein PrPC into a self-templating conformer PrPSc. Targeting PrPC with molecules that stabilize its native conformation has emerged as a promising approach for developing anti-prion therapies. A crystallographic structure of the globular domain of human PrPC showing a novel dimeric form and oligomeric arrangement has been reported, with molecular dynamics simulations used to explore its structural dynamics and stability, discussing potential implications for the conversion process.

Summary: The study reveals the core structure of prions and the templating surface formed by individual monomers at fibril ends. Comparison to another prion strain shows differences in fibril morphology. These findings provide insights into the propagation of prions.
