Irisin alleviates vascular calcification by inhibiting VSMC osteoblastic transformation and mitochondria dysfunction via AMPK/Drp1 signaling pathway in chronic kidney disease

Pei-Wen Wang et al.

Summary: The results of this study reveal for the first time that Irisin inhibits vascular calcification (VC) associated with chronic kidney disease (CKD). Irisin suppresses vascular smooth muscle cells (VSMCs) osteoblastic transformation and mitochondrial dysfunction via AMPK/Drp1 signaling.

Daprodustat Accelerates High Phosphate-Induced Calcification Through the Activation of HIF-1 Signaling

Andrea Toth et al.

Summary: The study found that DPD stabilized HIF-1α and HIF-2α in VSMCs and activated the HIF-1 pathway, leading to increased phosphate-induced calcification. Oral administration of DPD corrected anemia but increased aortic calcification in CKD mice, suggesting a need for long-term follow-up studies.

Multiple functions of pyruvate kinase M2 in various cell types

Yoon-Beom Lee et al.

Summary: PKM2, a key metabolic enzyme, plays multiple roles in cells including traditional metabolic functions, transcriptional regulation, and protein kinase activity. Its posttranslational modifications can alter cellular functions. PKM2 is highly expressed in cancer cells, embryonic cells, and astrocytes, while mature neurons predominantly express PKM1.

Hydroxyapatite nanoparticles promote mitochondrial-based pyroptosis via activating calcium homeostasis and redox imbalance in vascular smooth muscle cells

Yubin Xia et al.

Summary: This study comprehensively characterized the adverse biological effects and mechanism of action of hydroxyapatite nanoparticles (HAP) on vascular smooth muscle cells. It was found that HAP induces calcium homeostasis imbalance and oxidative stress, leading to cell death and enhancing vascular calcification. In addition, in vivo experiments demonstrated that HAP accumulates in the kidneys and causes kidney damage.

Summary: The redox balance of cells is crucial for providing a stable environment for biological macromolecules, and its imbalance is closely related to disease development. Precision redox strategies, emphasizing differences in redox status and proposing the 5R principle, are key for successful antioxidant pharmacology. The precise application of antioxidants is crucial for general health and should consider individual variations.

Summary: Vascular calcification refers to the deposition of calcium apatite within the vascular wall, which leads to vascular stiffening and loss of compliance. Mitochondrial dysfunction may play a pathogenic role in the process of vascular calcification, with SOD2 potentially exerting beneficial effects by attenuating mitochondrial reactive oxygen species. Additionally, SOD2 has been found to affect various cellular processes that contribute to the development of vascular calcification, suggesting its potential as a druggable target for future therapeutic strategies.
