Cwc27, associated with retinal degeneration, functions as a splicing factor in vivo

Renae Elaine Bertrand et al.

Summary: Previous studies have shown that CWC27 acts as a splicing factor in the B-act spliceosome complex, and CWC27 variants can cause autosomal recessive retinal degeneration. By utilizing the Cwc27(K338fs/K338fs) mouse model, it was demonstrated that Cwc27 is involved in splicing in the retina, leading to gene expression and splicing pattern changes that may contribute to disease mechanisms such as ER stress activation. This study provides new evidence for the role of CWC27 as a splicing factor in an in vivo context.

Specialization of the photoreceptor transcriptome by Srrm3-dependent microexons is required for outer segment maintenance and vision

Ludovica Ciampi et al.

Summary: Retinal photoreceptors have a unique transcriptomic profile, including distinct microexons and a microexon program regulated by Srrm3. Deletion of Srrm3 may lead to retinal diseases, highlighting the transcriptomic specialization and functionality of photoreceptors.

Integrative genome-wide analysis reveals EIF3A as a key downstream regulator of translational repressor protein Musashi 2 (MSI2)

Shilpita Karmakar et al.

Summary: This study identified the specific RNA binding partners of MSI2 using iCLIP and found that MSI2 regulation is not simply triggered by binding. The functional targets of MSI2 were found to have higher binding density and contain more 'UAG' motifs compared to nonproductive targets. Acute depletion of MSI2 only induced translational changes in a small fraction of transcripts, highlighting the challenges in defining functional targets of RBPs.

Local Translation Across Neural Development: A Focus on Radial Glial Cells, Axons, and Synaptogenesis

Manasi Agrawal et al.

Summary: Recent research has shown significant progress in understanding mRNA localization and local translation in axons and dendrites. It has been discovered that these processes are essential for neural development and play a crucial role in mediating structural and functional changes during synaptogenesis. Further studies are needed to explore the functional significance of mRNA localization and local translation in various stages of neural development.

The Musashi proteins MSI1 and MSI2 are required for photoreceptor morphogenesis and vision in mice

Jesse Sundar et al.

Summary: The Musashi family of RNA-binding proteins plays a critical role in the morphogenesis of terminally differentiated photoreceptor neurons in the retina. Loss of MSI1 and MSI2 in mice led to disrupted photoreceptor outer segment morphology and eventually neuronal degeneration, highlighting the importance of Musashi proteins in regulating key transcripts critical for photoreceptor function.
