Comparative chemical and biological hydrolytic stability of homologous esters and isosteres

Hygor M. R. de Souza et al.

Summary: This paper investigates the stability of homologous esters and isosteres in different media and reveals that the stability is related to carboxylesterase activity. Molecular modeling studies are conducted to design bioactive compounds or prodrugs with higher or lower metabolic stability.

Activation of benzoate model prodrugs by mycobacteria. Comparison with mammalian plasma and liver hydrolysis

Emilia Valente et al.

Summary: Linear alkyl benzoates show a parabolic dependence between log(k) and logP in all biological media, with a maximal value for the n-butyl chain in mycobacterial esterases. pKa(LG) seems to be the most important descriptor for mycobacterial esterases, while plasma esterases are sensitive to sigma* and pKa(LG), and liver esterases are influenced more by E-S(c). Lipophilicity weakly correlates with log (k) in all media, but becomes more important when considering mycobacterial activation selectivity.
