Enhanced anti-angiogenetic effect of transferrin receptor-mediated delivery of VEGF-trap in a glioblastoma mouse model

Summary: This study investigates the potential of enhancing the efficacy of anti-angiogenic antibody therapies for glioblastoma (GBM) by increasing drug concentration in the brain through transferrin receptor (TfR)-mediated delivery across the blood-brain barrier (BBB). The researchers identified an antibody that binds to the apical domain of the mouse TfR and engineered two bispecific antibodies fusing a vascular endothelial growth factor (VEGF)-Trap with the TfR-targeting antibody. The results showed that the engineered antibody significantly improved the brain distribution of VEGF-Trap and inhibited angiogenesis in a mouse model of GBM.
