Dendrimer-Based Nanogels for Cancer Nanomedicine Applications

Summary: Nanogels are nanosized superconstructs composed of amphiphilic or hydrophilic polymer networks, with unique physicochemical properties that make them suitable for various biomedical applications, particularly in cancer nanomedicine. Dendrimer-based NGs can be synthesized via different methods and used for drug delivery, gene delivery, photothermal therapy, combination therapy, and biological imaging-guided chemotherapy in cancer treatment. Future perspectives include designing new dendrimer-based NGs for different cancer nanomedicine applications.

Curing inflammatory diseases using phosphorous dendrimers

Summary: Different types of water-soluble phosphorous dendrimers with various biological properties, such as stimulating the immune system and displaying anti-inflammatory effects, have been synthesized. These dendrimers have shown potential in the treatment of chronic or acute inflammatory diseases and hematological cancers in animal models. Safety assessments have demonstrated their hematological safety both in mice and non-human primates.

Astodrimer sodium and bacterial vaginosis: a mini review

Summary: Bacterial vaginosis (BV) is a common vaginal infection that greatly affects the physical, emotional, sexual, and social lives of women of childbearing age. Astodrimer sodium gel is a novel non-antibiotic treatment for BV that prevents pathogenic bacteria from adhering to the vaginal wall and inhibits the formation of bacterial biofilms. Clinical trials have shown higher cure rates and lower recurrence rates in patients treated with astodrimer sodium compared to placebo.

The DendrisCHIP® Technology as a New, Rapid and Reliable Molecular Method for the Diagnosis of Osteoarticular Infections

Summary: In this study, a molecular diagnostic technology called DendrisCHIP (R) OA was reported for accurate identification of pathogen bacteria in osteoarticular infections. This technology combines the advantages of PCR and NGS while overcoming their limitations and constraints. Compared to microbial culture and NGS, DendrisCHIP (R) OA is able to detect a wider range of pathogens and provides faster diagnostic results.

Subcutaneous delivery of a dendrimer-BH3 mimetic improves lymphatic uptake and survival in lymphoma

Summary: This study investigates the impact of subcutaneous dosing on lymphatic exposure, pharmacokinetics, and efficacy of AZD0466. The results show that subcutaneous dosing leads to longer half-life, higher lymphatic transport rate, and improved tolerance. Additionally, higher doses of subcutaneous administration result in improved efficacy.

Summary: This study assessed the safety and absorption of Astodrimer sodium as a nasal spray for antiviral purposes. The results showed that Astodrimer sodium was well tolerated when applied to the nasal mucosa and did not cause systemic absorption. A novel BH3-mimetic, AZD0466, targeting BCL-XL and BCL-2 is effective in pre-clinical models of malignant pleural mesothelioma

Summary: The study found that the inhibitor AZD4320 targeting BCL-XL can effectively kill malignant pleural mesothelioma tumor cells, with enhanced effects when used in combination with AZD5991. A novel nanoparticle, AZD0466, showed similar effectiveness to standard chemotherapy drug Cisplatin in inhibiting tumor growth in mouse xenograft studies, while reducing thrombocytopenia associated with BCL-XL inhibition.

Summary: The study presents the design and development of AZD0466, a drug-dendrimer conjugate, and uses preclinical and mathematical models to determine the optimal release rate of the drug for maximal therapeutic index. Silencing of SARS-CoV-2 with modified siRNA-peptide dendrimer formulation

Summary: The study identified siR-7 as the most efficient siRNA targeting SARS-CoV-2 RNA-dependent RNA polymerase in inhibiting viral replication in vitro. Modification with LNAs and complexation with the designed peptide dendrimer enhanced the antiviral capacity of siR-7 in vitro. Inhalation of siR-7-EM/KK-46 led to significant reduction of virus titer and lung inflammation in vivo, demonstrating its potential as a therapeutic strategy for COVID-19 patients.
