Prevalence of polymorphisms in thiopurine metabolism and association with adverse outcomes: a South Asian region-specific systematic review and meta-analysis

Anuraag Jena et al.

Summary: In the South Asian population, NUDT15 polymorphisms are more prevalent than TPMT and are more frequently associated with adverse events. These findings suggest the importance of preemptive testing for NUDT15 among South Asian population and immigrants before starting thiopurines.

Genetic profiles of 103,106 individuals in the Taiwan Biobank provide insights into the health and history of Han Chinese

Chun-Yu Wei et al.

Summary: The Taiwan Biobank has collected genetic information from both high-coverage whole-genome sequencing and genome-wide SNP data of individuals with Han Chinese ancestry, revealing the full range of genetic variation in this population. The study found that some individuals carry mutations related to autosomal recessive diseases, cancer-predisposing genes, and variants affecting drug response, highlighting the potential for genetic testing to improve clinical care.

Hepatotoxicity reports in the FDA adverse event reporting system database: A comparison of drugs that cause injury via mitochondrial or other mechanisms

Payal Rana et al.

Summary: Drug-induced liver injury (DILI) is a significant cause of preclinical safety attrition and postmarket drug withdrawals, with mitochondrial toxicity playing a critical role, especially in idiosyncratic liver injury. Reports indicate that DILI involving mitochondrial mechanisms have a higher odds of occurrence compared to non-mitochondrial mechanisms, with the top drugs associated with mitochondrial toxicity having the highest risk values. Age and gender are also important factors influencing DILI risk, with older patients and females showing different patterns of liver injury reports.
