Calcific aortic valve disease: from molecular and cellular mechanisms to medical therapy

Summary: Calcific aortic valve disease is a prevalent condition with a disease continuum that leads to aortic stenosis and heart failure. Despite shared risk factors with atherosclerotic cardiovascular disease, the molecular and cellular mechanisms differ, requiring new targets for treatment. Completed phase II trials show potential for non-invasive pharmaceutical tactics targeting different mechanisms of CAVD pathogenesis.

Low levels of Gla-rich protein (GRP) and magnesium, along with high levels of phosphate, are associated with mitral and aortic valves calcification. The diagnostic value of GRP may be potentially useful in assessing cardiovascular risk in this population.
