Robust genome editing in adult vascular endothelium by nanoparticle delivery of CRISPR-Cas9 plasmid DNA

Xianming Zhang et al.

Summary: A newly developed nanoparticle-based delivery system has shown high efficiency in genome editing of vascular endothelial cells. This system can selectively decrease protein expression by 80%, achieving a phenotype similar to genetic knockout mice. Furthermore, it has the capability to edit multiple genes or perform both genome editing and transgene expression simultaneously, making it a powerful tool for gene therapy and cardiovascular research.

Polyurethane-polyurea hybrid nanocapsules as efficient delivery systems of anticancer Ir(III) metallodrugs

Joaquin Bonelli et al.

Summary: By encapsulating poorly soluble cyclometalated Ir(III) complexes using polyurethane-polyurea hybrid nanocapsules, their solubility is improved and the nanocapsules can enhance cellular uptake and cytotoxicity, leading to inhibition of tumor growth.

Recent advances in the redox-responsive drug delivery nanoplatforms: A chemical structure and physical property perspective

Shirin Mollazadeh et al.

Summary: Redox-responsive drug delivery nanoplatforms address the challenges in conventional drug delivery and enable targeted delivery, widely used in designing nanoparticles. By cleaving diselenide and disulfide bonds, controlled loading, delivery, and release of poorly water-soluble anticancer drugs at tumor site can be achieved.

Multi-Smart and Scalable Bioligands-Free Nanomedical Platform for Intratumorally Targeted Tambjamine Delivery, a Difficult to Administrate Highly Cytotoxic Drug

Marta Perez-Hernandez et al.

Summary: The study developed a novel drug delivery system based on nanocapsules, which could enhance the concentration and selectivity of anticancer drugs at tumor sites, showing increased cytotoxicity in acidic conditions. Results indicate that the system effectively reached tumor sites in mice, reduced systemic toxicity of free drugs, and maintained antitumor activity.

Universal quenching of common fluorescent probes by water and alcohols

Jimmy Maillard et al.

Summary: This study demonstrates that water acts as a classic fluorescence quencher for organic fluorophores, reducing their fluorescence quantum yield and lifetime significantly. The fluorescence quenching is found to be more efficient for red-emitting probes and is attributed to high-energy vibrations of the solvent. Understanding this solvent-assisted quenching mechanism could lead to the rational design of brighter fluorescent probes by protecting organic fluorophores from the solvent.
