Gene Amplification of CYP518: a New Mechanism of Resistance to Azole Compounds in Trichophyton indotineae

Summary: Trichophyton indotineae strains resistant to itraconazole and voriconazole were found to be associated with overexpression of TinCYP518 due to additional copies of this gene. RNA interference of TinCYP518 restored drug susceptibility in resistant strains, while overexpression of TinCYP518 conferred resistance in susceptible strains of T. indotineae.

Use of real time multiplex PCR for the diagnosis of dermatophytes onychomycosis in patients with empirical antifungal treatments

Summary: This study evaluated the DermaGenius (DG) multiplex kit for detecting and identifying dermatophytes from nail samples of untreated and treated patients with a clinical suspicion of onychomycosis. The results showed that DG had a high sensitivity and specificity in diagnosing onychomycosis, especially in patients who had undergone antifungal treatments without clinical improvement. The positive results of cultural exams were significantly lower in treated patients compared to untreated patients.
