From environmental adaptation to host survival: Attributes that mediate pathogenicity of Candida auris

Stefanie Allert et al.

Summary: Candida auris is a newly emerged species causing invasive fungal infections with high rates of treatment failures. Our study revealed that C. auris is efficiently killed in human blood, but exhibits unique patterns of immune cell association, survival rates, and cytokine induction compared to other Candida species. The transcriptional adaptations of C. auris, its increased stress resistance and long-term environmental survival likely contribute to its high risk of contamination and distribution in healthcare settings.

Comparative Transcriptomics Reveal Possible Mechanisms of Amphotericin B Resistance in Candida auris

Raju Shivarathri et al.

Summary: This study suggests that membrane lipid alterations and chromatin modifications play critical roles in adaptive AmB resistance in clinical C. auris isolates. Transcriptomic analysis revealed differential gene expression profiles between AmB-resistant and AmB-sensitive isolates, highlighting genes involved in lipid and ergosterol biosynthesis, adhesion, drug transport, and chromatin remodeling. Additionally, AmB-resistant strains showed increased adhesion, reduced lipid membrane permeability, and hyper-resistance to cell wall perturbing agents. The phosphorylation of Mkc1 cell integrity MAP kinase was also elevated upon AmB treatment.

Biomarkers of caspofungin resistance in Candida albicans isolates: A proteomic approach

Giuseppe Buda De Cesare et al.

Summary: This study reveals that echinocandin-resistant clinical isolates of Candida albicans exhibit different cell surface characteristics compared to sensitive isolates and show a different response to echinocandin drugs. These findings suggest that drug resistance may involve alterations in cell wall architecture and drug response.

Insight Into the Properties and Immunoregulatory Effect of Extracellular Vesicles Produced by Candida glabrata, Candida parapsilosis, and Candida tropicalis Biofilms

Kamila Kulig et al.

Summary: EVs produced by non-albicans Candida species have a proinflammatory effect on host immune cells, influencing cytokine production and participating in fungal biofilm resistance to drugs. This highlights the importance of EVs not only in the physiology of non-albicans Candida species, but also in the pathogenesis of infections associated with fungal biofilm production.

Cellular and Extracellular Vesicle RNA Analysis in the Global Threat Fungus Candida auris

Isadora F. Munhoz da Rocha et al.

Summary: This study identified RNA content in extracellular vesicles (EVs) and cellular small RNA of Candida auris under normal growth conditions and after treatment with the antifungal drug caspofungin. The data set provides insights into fungal biology and may aid in understanding antifungal treatment, gene expression, and EV composition. This study contributes to the exploration of small RNA content in the fungal kingdom and sheds light on the mechanisms of molecules directed to fungal EVs.

Transcriptional and translational landscape of Candida auris in response to caspofungin

Daniel Zamith-Miranda et al.

Summary: Candida auris has emerged as a serious global threat due to its high resistance to conventional antifungal medications. This study treated two strains of C. auris with caspofungin and found that it caused morphological alterations, changes in cell wall composition, and upregulation of genes related to cell wall synthesis. These results provide valuable insights into the biology of C. auris and the antifungal activity of caspofungin, identifying potential cellular targets for the development of novel antifungals.

Mechanism of cargo sorting into small extracellular vesicles

Yiwen Chen et al.

Summary: Extracellular vesicles are membranous structures released by cells to transport biomolecules and signaling molecules between cells, with small sEVs and larger EVs being the two main types categorized by size. These vesicles are no longer considered waste products, but rather regulators of intercellular communication with specific cargos sorted into them through different mechanisms.
