RhoA/ROCK2 signalling is enhanced by PDGF-AA in fibro-adipogenic progenitor cells: implications for Duchenne muscular dystrophy

Summary: The lack of dystrophin expression in Duchenne muscular dystrophy (DMD) induces muscle fibre and replacement by fibro-adipose tissue. Our study found that PDGF-AA promotes the activation of RhoA pathway in FAPs from DMD patients, leading to proliferation, migration, and actin reorganization, which represents the beginning of the fibrotic process. Inhibition of the RhoA pathway could be considered as a potential therapeutic target for muscle fibrosis in patients with muscular dystrophies.

P2X7 Receptor Antagonist Reduces Fibrosis and Inflammation in a Mouse Model of Alpha-Sarcoglycan Muscular Dystrophy

Summary: In this study, the therapeutic effectiveness of a selective P2X7 purinoreceptor antagonist, A438079, was evaluated in limb-girdle muscular dystrophy R3. The results showed that the P2X7 antagonist improved clinical parameters, reduced muscle inflammation and fibrosis, and upregulated immunosuppressive regulatory T cells.

Clinical and genetic spectrum of a large cohort of patients with δ-sarcoglycan muscular dystrophy

Summary: Sarcoglycanopathies are a group of genetic muscular diseases, with delta-sarcoglycanopathy being the rarest subtype. This study retrospectively analyzed a large cohort of delta-sarcoglycanopathy patients and found that the disease is severe and rapidly progressive, predominantly affecting limb muscles. Similar to other forms of sarcoglycanopathies, the abundance of protein on muscle biopsy inversely correlates with disease severity and progression rate.

Summary: Duchenne muscular dystrophy and other muscular dystrophies are characterized by progressive muscle degeneration, inflammation, and fibrosis. LTBP4, an extracellular matrix protein found in muscle, has two different alleles - a protective form with increased sequestration of latent TGF beta and reduced muscle fibrosis, and a deleterious form that promotes release of latent TGF-beta. A monoclonal human anti-LTBP4 antibody directed toward LTBP4's hinge region has been shown to stabilize muscle membrane, reduce proteolytic cleavage of LTBP4 protein, protect against muscle force loss, reduce fibrosis, and enhance muscle force production, making it a potential treatment for muscular dystrophy.
