The Effect of Growth Stage and Isolation Method on Properties of ClearColi™ Outer Membrane Vesicles (OMVs)

Summary: Outer membrane vesicles (OMVs), derived from gram-negative bacteria, play important roles in various physiological functions. This study aimed to determine the appropriate growth phase and isolation method for OMV separation from an endotoxin-free strain of E. coli. It was found that combining protein precipitation by ammonium sulfate (AS) with ultracentrifugation (UC) resulted in the highest yield of OMVs, although slightly larger and more heterogeneous OMVs were obtained using a precipitation-based exosome isolation kit.

Delivery of functional exogenous proteins by plant-derived vesicles to human cells in vitro

Summary: Plant-derived extracellular vesicles (EVs) are gaining attention as promising carriers for delivering exogenous bioactive molecules to human cells. In this study, grapefruit EVs were isolated and characterized, showing efficient delivery of proteins to human cells and significant uptake by organs in mice. These findings suggest that native plant EVs could be safe and effective carriers for exogenous proteins.

A novel decoy strategy for polymyxin resistance in Acinetobacter baumannii

Summary: The modification of outer membrane charge induced by polymyxin B (PMB) in Acinetobacter baumannii leads to PMB-resistant variants and clinical isolates producing outer membrane vesicles (OMVs). Upregulation of the pmr operon and decreased membrane-linkage proteins result in enhanced biofilm formation and protection of susceptible bacteria from PMB, suggesting OMVs function as decoys against PMB. OMVs also protect the microbial community against PMB treatment in anaerobic conditions, indicating their potential clinical significance in antibiotic resistance.
