Itaconate attenuates osteoarthritis by inhibiting STING/NF-κB axis in chondrocytes and promoting M2 polarization in macrophages

Summary: This study found that exogenous supplementation of itaconate inhibits inflammation and senescence of chondrocytes through the Nrf2/STING/NF-kappa B axis, reducing the progression of osteoarthritis. Itaconate also regulates the polarization of macrophages, reducing apoptosis of chondrocytes. In vivo, intra-articular injection of itaconate reduces cartilage degradation and synovial membrane inflammation in a mouse model of osteoarthritis.

Furthermore, the in situ structural transformation also allows for noninvasive monitoring of therapeutic effects as a photoacoustic imaging contrast agent.

Summary: This study demonstrates the design and development of a smart anti-oxidative agent with dual-responsive hybrid micelles for photoacoustic imaging-guided knee OA therapy. The resulting micelles exhibit long-term controlled-release behavior under endogenous ROS stimuli and short-burst release triggered by exogenous NIR light, showing excellent free radical scavenging capacity and protective effects on chondrocytes. Additionally, the micelles effectively provide PA imaging for therapy adjustment and protect OA from inflammation via TLR-2/MAPK/Akt signaling.

The nanodrug also inhibits cartilage degradation, bone erosion, and proinflammatory cytokines, showcasing potential for treating various inflammatory diseases.
