Untuned antiviral immunity in COVID-19 revealed by temporal type I/III interferon patterns and flu comparison

Ioanna-Evdokia Galani et al.

Summary: Andreakos and colleagues conducted a study comparing patients with COVID-19 and influenza, finding that COVID-19 patients exhibited a dysregulated interferon response with diminished and delayed interferon production compared to flu patients. In COVID-19 patients, pro-inflammatory cytokines were produced before interferons and persisted longer, leading to worsened clinical outcomes.

Interleukin-3 is a predictive marker for severity and outcome during SARS-CoV-2 infections

Alan Benard et al.

Summary: Researchers identify IL-3 as an independent prognostic marker for the outcome of SARS-CoV-2 infections, with low IL-3 levels being associated with increased severity, viral load, and mortality. Additionally, patients with severe COVID-19 have reduced levels of circulating plasmacytoid dendritic cells compared to non-severe cases.

Monocyte and dendritic cell defects in COVID-19

Andreas Wack

Summary: A new study compares severe and moderate cases of COVID-19 and finds a link between defects in viral sensing, interferon and antigen presentation pathways, as well as upregulated apoptosis and inflammatory pathways, to high COVID-19 severity.

Cellular and Humoral Immune Responses in Covid-19 and Immunotherapeutic Approaches

Amal Hasan et al.

Summary: Severe Covid-19 is attributed to a dysfunctional innate immune response with delayed and/or deficient type I interferon response, coupled with an exaggerated and/or dysfunctional adaptive immunity. Differences in T-cell and B-cell responses, as well as neutralizing antibody responses, have been identified in severe disease compared to mild cases. Preexisting autoantibodies against type I interferon have been described as a major cause of severe/critical disease. Additionally, factors such as prior vaccine exposure, trained innate immunity, cross-reactive immunity, and serological immune imprinting may all contribute towards disease severity and outcome.
