Summary: Despite concerns over the use of fetal bovine serum (FBS), it is still widely used in cell culture media for isolating and expanding mesenchymal stromal cells (MSCs). Research on alternative supplements and serum-free media has been conducted, and while none of them can completely replace FBS, short-term or transitional use of FBS-free media can be equally effective.

Summary: The study demonstrated that IL-1 beta can enhance the immunoregulatory activity of MSC induced by TNF-alpha/IFN-gamma. TNF-alpha and IL-1 beta increased the expression of IFN-gamma receptor via NF-kappa Beta signaling, leading to activation of STAT5 and p38-MAPK signaling, resulting in increased IL-8 release and recruitment of polymorphonuclear granulocytes. These findings suggest therapeutic potential for exploiting MSC responses to multi-cytokine priming regimens in fine-tuning inflammatory activity in tissues.

Effects of continuous passage on the immunomodulatory properties of equine bone marrow-derived mesenchymal stem cells in vitro

Summary: The study investigated the impact of continuous passage on the immunomodulatory properties of equine bone marrow-derived MSCs. Results showed that T cell proliferation was suppressed by BM-MSCs from all passages, with no significant differences between passage numbers. All passages of BM-MSCs increased cytokine synthesis following stimulation with IFN-gamma. However, higher passage numbers exhibited significant differences in the production of certain cytokines compared to lower passage numbers.

Summary: Research shows that there are significant differences between MSCs prepared with FBS and MSCs prepared without FBS, with FBS-prepared MSCs causing inflammation and antibody-mediated cytotoxicity. Additionally, the efficacy of FBS-prepared MSCs in treatment is reduced. Therefore, it is recommended to no longer use FBS in the preparation of MSCs.
