Impact of Albumin Pre-Coating on Gold Nanoparticles Uptake at Single-Cell Level

Summary: This study investigated the impact of bovine serum albumin (BSA) pre-coating on the cellular uptake of nanoparticles (NPs). The results showed that the well-dispersed NPs-BSA complexes were more stable in the culture medium and were less taken up by cells. Additionally, the settled NPs agglomerates played a significant role in determining the average uptake at the population level.

Iron Oxide Nanoparticle Coatings Dictate Cell Outcomes Despite the Influence of Protein Coronas

Summary: In the study, it was found that different coatings did not affect the composition of the protein corona on iron oxide nanoparticles. Enzymes with hydrolase activity in the corona may explain the degradation of coatings when in contact with biological media. Coatings primarily determine the endocytic pathways used for nanoparticle internalization by cells.

Composition of Intracellular Protein Corona around Nanoparticles during Internalization

Summary: This study used a PFA cross-linking strategy to stabilize the composition of protein corona around intracellular nanoparticles, revealing that the corona is mainly composed of cell-derived proteins and undergoes significant changes over time during internalization. The results indicate that AuNPs are predominantly endocytosed through a clathrin-mediated pathway into an endolysosomal pathway.

Summary: Natural photosensitizers, like flavins, have excellent toxicity profiles and activating energy ranges compatible with biological systems. Flavin-rich nanoparticles show promise in photodynamic therapies and drug delivery with their improved pharmacokinetics and reduced photobleaching. The design of riboflavin-rich nanoparticles with tunable morphology and predictable self-assembly can be guided by coarse-grained simulations of the self-assembling process.

Research shows that the protein corona has different impacts on receptor targeting, lysosomal escape, and protein transcytosis in BBB. After crossing the BBB, transferrin retains its targeting specificity towards brain tumor cells.

Summary: This study demonstrated the use of modified PLGA-CSRf nanoparticles as both drug delivery ligands and photodynamic therapy agents for TNBC cells. The biocompatibility of the nanoparticles was confirmed in both breast cancer and normal breast cells, with a significant increase in cellular uptake in cancer cells compared to normal cells. The results suggest that riboflavin can enhance the delivery of PLGA nanoparticles to TNBC cells and improve the therapeutic effects of combined drug delivery and photodynamic therapy.
