An integrated multi-omics approach revealed the regulation of melatonin on age-dependent mitochondrial function impair and lipid dyshomeostasis in mice hippocampus

Xiaowen Jiang et al.

Summary: Melatonin can improve mitochondrial dysfunction associated with aging by removing active oxygen and inhibiting lipid peroxidation. It can also regulate mitochondrial electron transport and lipid metabolism in the hippocampus, and improve damaged mitochondria and morphology. Chronic supplementation of low-dose melatonin reverses age-dependent up-regulation of long-chain unsaturated lipids.

Design, synthesis and biological evaluation of harmine derivatives as potent GSK-3β/DYRK1A dual inhibitors for the treatment of Alzheimer's disease

Wenwu Liu et al.

Summary: Harmine derivatives were designed, synthesized, and evaluated as dual GSK-3 beta/DYRK1A inhibitors, showing promising activity against both targets. Compound ZDWX-25 exhibited potent inhibitory effects on GSK-3 beta and DYRK1A, with IC50 values of 71 and 103 nM, respectively. These compounds have the potential to be promising candidates for Alzheimer's disease treatment.

The State of The Art on Acetylcholinesterase Inhibitors in the Treatment of Alzheimer's Disease

Immacolata Vecchio et al.

Summary: Alzheimer's disease is a chronic disabling disease that affects the central nervous system, leading to cognitive decline and language disorders. One of the causes of AD is the decrease of acetylcholine levels in the brain. Acetylcholinesterase inhibitors have been developed to counteract the progression of the disease, with drugs like donepezil and galantamine showing promising clinical benefits.
