Systematic comparison of activity and mechanism of antimicrobial peptides against nosocomial pathogens

Bruce Lin et al.

Summary: The World Health Organisation has identified several multi-drug resistant nosocomial bacterial pathogens as a significant threat to human health. This study compares the antimicrobial activities of different antimicrobial peptides (AMPs) against these pathogens and investigates their modes of action. Pardaxin and MSI-78 show the most pronounced antimicrobial activity and their strong membrane interaction and structure contribute to their potency. The study provides guidance for the design of therapeutics against multi-drug resistant pathogens.

Structure of neuroendocrine regulatory peptide-2 in membrane-mimicking environments

One-Sung Park et al.

Summary: NERP-2, derived from the VGF neuropeptide precursor, exhibits a partially helical structure in buffer solutions, while showing a more stable helical structure in HFIP or DPC micelle. The NMR spectroscopy determined its 3D structure in both types of solutions, showing predominantly helical structure in HFIP and partial helical structure in DPC solutions. The amphipathic nature of the helical structure suggests potential structural changes in NERP-2 upon receptor binding.

Local rigidification and possible coacervation of the Escherichia coli DNA by cationic nylon-3 polymers

Yanyu Zhu et al.

Summary: Synthetic cationic random nylon-3 polymers show potential as inexpensive antimicrobial agents, affecting bacterial cells similarly to natural antimicrobial peptides. The study highlights the importance of charge density and molecular length in governing the antimicrobial effects, supporting a model where agents with sufficient cationic charge form a dense network of electrostatic pseudo-crosslinks locally rigidifying DNA in bacterial cells. Additionally, the longer polymers cause gradual coalescence of nucleoid lobes in bacterial cells, possibly through DNA coacervation similar to liquid droplet coacervates observed in eukaryotic cells.

Beyond structural models for the mode of action: How natural antimicrobial peptides affect lipid transport

Josefine Eilso Nielsen et al.

Summary: By combining state-of-the-art small angle X-ray/neutron scattering techniques, this study found that various natural antimicrobial peptides accelerate essential lipid transport processes without altering the overall structure of lipid membranes, potentially triggering lethal signaling processes and enhancing ion transport.

Expression and purification of the native C-amidated antimicrobial peptide maculatin 1.1

Shiying Zhu et al.

Summary: A recombinant expression protocol was reported to produce isotopically enriched Mac1 with native N and C termini amidation achieved in a one-pot reaction. The protocol yielded native, uniformly N-15-labeled Mac1 with identical structure and activity to peptide obtained by solid-phase peptide synthesis.

Antibiofilm activity of host defence peptides: complexity provides opportunities

Robert E. W. Hancock et al.

Summary: This review discusses how the complex functional attributes of host defence peptides offer opportunities for antimicrobial therapeutics, with a focus on their emerging antibiofilm properties. The mechanisms of action of antibiofilm peptides are compared with antimicrobial peptides. Additionally, obstacles to translating candidate peptides into therapeutics and potential solutions are discussed.
