Chromosomes polymorphisms of Sichuan wheat cultivars displayed by ND-FISH landmarks

Zhenglei Hu et al.

Summary: This study utilized novel oligonucleotide probes to investigate 166 common wheat cultivars bred from the hometown of Chinese Spring, revealing different chromosome variations in the A, B, and D genome chromosomes. Based on FISH karyotyping, the wheat cultivars were classified into eight clusters corresponding to breeding entities.

Centromere-Specific Single-Copy Sequences of Secale Species

Zijin Pan et al.

Summary: Single-copy FISH analysis is a useful tool for locating specific sequences on chromosomes. Centromeric single-copy sequences can be used to determine centromere position and reveal differences between centromeres. Nine centromeric single-copy sequences were cloned from Kustro (Secale cereale L.) and were found to be located in the centromeric regions of rye chromosomes. High sequence similarity was observed among different Secale species. The sequences are useful for determining centromere location in rye chromosomes and have the potential to uncover accurate structural differences among wheat-rye centric fusion translocation chromosomes.

Evolution and origin of bread wheat

Avraham A. Levy et al.

Summary: Bread wheat is a young hexaploid species that formed through hybridization between a domesticated tetraploid progenitor and a diploid donor around 8,500-9,000 years ago. Its global expansion was enabled by its polyploidy, which allowed for genetic novelty and economic appeal. The origins of its subgenomes are still debated, and efforts are needed to protect and utilize the genetic diversity of wheat for food security.

Universal fluorescence in situ hybridization (FISH) protocol for mapping repetitive DNAs in insects and other arthropods

Diogo Cavalcanti Cabral-de-Mello et al.

Summary: Repetitive DNAs are a large portion of eukaryote genomes, posing challenges in genome assembly due to the similarity between these repeats. The FISH technique is powerful for physical mapping of sequences on chromosomes, but there is a need for a universal and affordable protocol. Testing a FISH protocol on various insects and arthropods showed high quality results, indicating its potential utility in studying the evolution and role of repetitive DNA in arthropod genomes.

FISH landmarks reflecting meiotic recombination and structural alterations of chromosomes in wheat (Triticum aestivum L.)

Yang Zou et al.

Summary: The study identified the impact of tandem repeat composition on meiotic recombination in common wheat by using specific probe detection methods on the long arm of wheat 5A chromosome. This provides an important research tool for wheat breeding programs.
