Re-Use of Caco-2 Monolayers in Permeability Assays-Validation Regarding Cell Monolayer Integrity

Results showed that manipulation during assays may affect permeability measurements, leading to P-app values dependent on sampling duration. Summary: This study characterized the lipidome and lipid/protein ratio of membranes from High Five™ insect cells overexpressing mouse P-glycoprotein, providing insight into the lipid environment surrounding P-glycoprotein and its properties. The membranes also contained negatively charged lipids such as phosphatidylserines, phosphatidylinositols, and phosphatidylglycerols.
