Striatal neurons expressing dopamine D1 receptor promote wakefulness in mice

Hui Dong et al.

Summary: D1 receptor neurons within the dorsal striatum play a significant role in promoting wakefulness by integrating corticostriatal, thalamostriatal, and nigrostriatal projections, and are synchronized with upstream structures during sleep-wake transitions.

Narcolepsy genetic marker HLA DQB1*06:02 and excessive daytime sleepiness in Parkinson disease patients treated with dopaminergic agents

Octavian Adam et al.

Summary: This study aimed to investigate the association between the HLA risk allele DQB1*0602 and excessive daytime sleepiness (EDS) in patients with Parkinson's disease (PD). The results showed that PD patients with the DQB1*0602 allele were more likely to experience EDS and inappropriate sleep during activities that require sustained alertness. Genetic vulnerability may play a role in explaining the risk of EDS in PD patients.

The Dopamine D1 Receptor Positive Allosteric Modulator Mevidalen (LY3154207) Enhances Wakefulness in the Humanized D1 Mouse and in Sleep-Deprived Healthy Male Volunteers[S]

Andrew P. McCarthy et al.

Summary: This study provides the first translational evidence of the effects of a selective dopamine receptor 1 positive allosteric modulator (D1PAM) on sleep and wakefulness in both humanized dopamine receptor 1 mice and sleep-deprived healthy male volunteers. The findings suggest that sleep-wake activity can serve as a translational biomarker for D1PAM and that D1PAMs may have therapeutic potential for sleep disorders and excessive daytime sleepiness associated with neurodegenerative disorders.

Selegiline reduces daytime sleepiness in patients with Parkinson's disease

Marco Gallazzi et al.

Summary: The study found that the use of selegiline improved excessive daytime sleepiness in Parkinson's disease patients significantly, but did not change the evaluation of disease severity. Therefore, selegiline may be a valuable add-on therapy to reduce daytime somnolence in PD patients.

Summary: This article reviews the pathophysiology and clinical evaluation and management of excessive daytime sleepiness (EDS) in patients with obstructive sleep apnea (OSA), including the mechanisms underlying EDS in CPAP-treated patients and potential pharmacotherapy options for those who experience residual EDS despite primary OSA therapy. The research highlights the importance of further studies on the effects of wake-promoting treatments on cognition in these patients and individual or disorder-specific responses.
