Comparison of chemometric strategies for potential exposure marker discovery and false-positive reduction in untargeted metabolomics: application to the serum analysis by LC-HRMS after intake of Vaccinium fruit supplements

Lapo Renai et al.

Summary: This study presents different chemometric selection workflows for the discovery of potential exposure markers (PEMs) in untargeted LC-HRMS metabolomics, aiming to identify statistically significant PEMs using multivariate or univariate parametric or non-parametric data analysis methods. The study successfully identified a restricted group of significant features and annotated several metabolites associated with nutrition intake. Additionally, the analysis of longitudinal data revealed significant PEMs that can discriminate between different interventions.

Role of dietary polyphenols on gut microbiota, their metabolites and health benefits

S. Mithul Aravind et al.

Summary: In the past two decades, there has been intensive research on the beneficial health roles of dietary polyphenols in preventing oxidative stress related chronic diseases. As our understanding of the role of gut microbiota advances, the need to examine the prebiotic role of dietary polyphenols has emerged. Further research should focus on the importance of human trials and the links of polyphenols-gut microbiota-nerve-brain to unlock the mechanisms behind the observed benefits of dietary polyphenols.

Feature-based molecular networking for identification of organic micropollutants including metabolites by non-target analysis applied to riverbank filtration

Daniela Oberleitner et al.

Summary: This study demonstrates how non-target liquid chromatography (LC)-high-resolution tandem mass spectrometry (MS/MS) and the feature-based molecular networking (FBMN) workflow provide insight into water samples from four riverbank filtration sites with different redox conditions. Forty-three compounds were annotated, and four additional transformation products (TP) were successfully identified. Sartans and their TP were investigated regarding their removal behavior under different redox conditions and seasons for the first time. The study shows the power of FBMN in identifying previously unknown or unexpected compounds and their TP in water samples.

Advances in decomposing complex metabolite mixtures using substructure- and network-based computational metabolomics approaches

Mehdi A. Beniddir et al.

Summary: This review highlights the impact of computational metabolome mining tools on untargeted metabolomics analyses, providing a pathway to decompose complex metabolite mixtures into substructures and chemical class information. The tools discussed aim at processing liquid chromatography coupled to mass spectrometry fragmentation data, with a focus on substructure identification and chemical compound annotation. The technical advances outlined in this review have the potential to transform spectral data into knowledge that can address biological questions in various metabolomics disciplines.
