Harnessing microbial wealth for lignocellulose biomass valorization through secretomics: a review

Summary: The recalcitrance of lignocellulosic biomass poses challenges for its industrial-scale utilization, however, utilizing microbes for biomass degradation presents a promising solution. Through advancements in proteomics and mass spectrometry, researchers are able to identify key enzymes involved in lignocellulose deconstruction and develop enzyme cocktails for efficient biomass valorization. This review highlights the importance of secretomics in understanding microbial degradation of lignocelluloses and the potential in constructing synthetic microbial consortia for biomass conversion.

Screening and identifying microorganisms with feruloyl esterase activity in Chinese sesame-flavour baijiu fermentation materials (Jiupei)

Summary: In this study, microorganisms with feruloyl esterase activity were screened in the fermented raw materials of baijiu. Four different microorganisms with such activity were identified, and one strain, named J1, exhibited high feruloyl esterase activity. The strain J1, belonging to the genus Acetobacter, was found to have the closest relationship with Acetobacter fabarum and Acetobacter lovaniensis. The optimal conditions for the highest feruloyl esterase activity production by J1 were determined to be an initial fermentation pH of 7.0 at 30 degrees C, a 10% inoculation volume, and a 24-hour culturing period.

Summary: Feruloyl esterase (FAE), which catalyzes the hydrolysis of esterified sugar to release ferulic acid, was isolated from humus soil and heterogeneously expressed in Bacillus subtilis. The alkaline FAE showed optimal activity under alkaline pH, and when combined with other enzymes, it effectively degraded waste biomass and produced high-added value ferulic acid.
