Poricoic acid A (PAA) inhibits T-cell acute lymphoblastic leukemia through inducing autophagic cell death and ferroptosis

Summary: The study found that poricoic acid A (PAA) can inhibit the cell growth of T-cell acute lymphoblastic leukemia (T-ALL) and induce apoptosis. The mechanisms of action involve mitochondrial dysfunction and increased production of reactive oxygen species (ROS). Furthermore, PAA can suppress T-ALL growth by regulating autophagy and ferroptosis pathways. In vivo experiments confirmed the effectiveness of PAA in inhibiting T-ALL tumor growth without detectable toxicity.

MiR-376a-3p increases cell apoptosis in acute myeloid leukemia by targeting MT1X

Summary: Metallothionein1X (MT1X) is highly expressed in acute myeloid leukemia (AML) and acts as a candidate prognostic indicator. Inhibiting MT1X can suppress AML cell proliferation, induce apoptosis, and inhibit nuclear factor-kappa B (NF-kappa B) signaling. MiR-376a-3p negatively regulates MT1X, suppressing AML cell proliferation and inducing apoptosis. These findings suggest that elevated MT1X expression may contribute to AML progression and indicate that the miR-376a/MT1X axis could be a potential therapeutic target for AML patients.

mTOR inhibition downregulates glucose-6-phosphate dehydrogenase and induces ROS-dependent death in T-cell acute lymphoblastic leukemia cells

Summary: mTOR activation is associated with resistance to GC-based chemotherapy in T-ALL. In this study, it was found that the mTOR inhibitor everolimus increased ROS levels, activated NRF2, induced mitochondrial inner membrane depolarization and apoptosis of T-ALL cells. The combination of everolimus and dexamethasone had a synergistic effect on killing T-ALL cells.

N-acetylcysteine Can Induce Massive Oxidative Stress, Resulting in Cell Death with Apoptotic Features in Human Leukemia Cells

Summary: N-acetylcysteine (NAC) can induce ROS production in HL-60 and U937 cells, with cytotoxicity depending on the expression of ROS-modulating enzymes. In HL-60 cells, NAC induces cell death through NOX2, SOD1, SOD3, and MPO, while in U937 cells, lower expression levels of these enzymes lead to weaker cytotoxicity. The mode of cell death induced by NAC treatment in both cell lines is primarily apoptotic.
