Integrin-linked kinase (ILK): the known vs. the unknown and perspectives

Agata Gorska et al.

Summary: Integrin-linked kinase (ILK) plays a crucial role in important cellular processes, such as cell adhesion, cell growth, and systemic changes. However, the exact mechanisms and functions of ILK are still not fully understood. Despite the existence of multiple ILK isoforms, research tends to focus on the main isoform, while the functional differences of other isoforms are still unclear. ILK activity is regulated at multiple levels, but the functions of most modifications are unknown.

Endothelial cell-derived tetrahydrobiopterin prevents aortic valve calcification

Zongtao Liu et al.

Summary: This study found that the levels of BH4 in the circulation and aortic valve tissue were significantly decreased in patients with aortic valve calcification. Endothelial cell-specific deficiency of GCH1 contributed to the development of aortic valve calcification. Oxidized LDL induced osteoblastic differentiation of valvular interstitial cells, which was inhibited by BH4 supplementation. Generation of peroxynitrite and tyrosine nitration of DRP1 contributed to aortic valve calcification. Folic acid attenuated hypercholesterolemia-induced aortic valve calcification by increasing DHFR levels.

Calcific aortic valve disease: from molecular and cellular mechanisms to medical therapy

Simon Kraler et al.

Summary: Calcific aortic valve disease is a prevalent condition with a disease continuum that leads to aortic stenosis and heart failure. Despite shared risk factors with atherosclerotic cardiovascular disease, the molecular and cellular mechanisms differ, requiring new targets for treatment. Completed phase II trials show potential for non-invasive pharmaceutical tactics targeting different mechanisms of CAVD pathogenesis.

Transcription factor Sp2 promotes TGFB-mediated interstitial cell osteogenic differentiation in bicuspid aortic valves through a SMAD-dependent pathway

Rui Zheng et al.

Summary: This study found that the transcription factor Sp2 is expressed at low levels in valvular interstitial cells (VICs) from patients with bicuspid aortic valve (BAV), which negatively impacts the expression of microRNA-195-5p (miR-195-5p) by binding its promoter region. This partially promotes calcification through a SMAD-dependent pathway.

TGF-β2 and TGF-β1 differentially regulate the odontogenic and osteogenic differentiation of mesenchymal stem cells

Jingzhi Li et al.

Summary: The study found that Transforming Growth Factor-beta 2 (TGF-beta 2) promotes odontogenic differentiation and attenuates osteogenic differentiation, while Transforming Growth Factor-beta 1 (TGF-beta 1) plays an important role in regulating osteogenic differentiation and has complex effects on odontogenic differentiation.

Endothelial to Mesenchymal Transition: An Insight in Atherosclerosis

Qingyan Huang et al.

Summary: Atherosclerosis, a major cardiovascular disease leading to high morbidity and mortality worldwide, is associated with endothelial to mesenchymal transition (EndMT) which contributes to plaque formation and may be induced by risk factors for atherosclerosis. Understanding the molecular mechanisms underlying EndMT in atherosclerosis could potentially offer insights for reversing this condition.

Summary: The study utilized metabolomics profiling to analyze CAVD and AS, revealing a 30 metabolic profile that can distinguish AS patients from healthy controls, with metabolites related to nitric oxide metabolism and inflammation. Additionally, the experiment indicated that these metabolites may be linked to left ventricular remodeling and serve as potential blood biomarkers for AS diagnosis and timing of intervention.

POSTN promotes proliferation and epithelial-mesenchymal transition in renal cell carcinoma through ILK/AKT/mTOR pathway

Yuan-yuan Jia et al.

Summary: This study focused on investigating the detailed mechanisms of POSTN overexpression in RCC invasion and migration. The results revealed that POSTN overexpression could promote EMT in RCC cells through the IKL/AKT/mTOR pathway, while POSTN knockdown could suppress EMT and inhibit cell proliferation and migration. Furthermore, higher POSTN expression was found to promote angiogenesis in an RCC xenograft tumor in vivo via activating the IKL /AKT/mTOR pathway.
