Putative role of STING-mitochondria associated membrane crosstalk in immunity

Chenyu Xue et al.

Summary: Stimulator of interferon genes (STING) is a crucial regulator of innate immunity that recognizes intracellular DNA. Recent research has revealed that STING has diverse cell-specific immune functions in various pathological conditions. It has been discovered that STING's unique location on the mitochondria-associated membrane enables it to translate multiple signals into distinct cellular responses through mitochondria-ER communication.

Multi-mtDNA Variants May Be a Factor Contributing to Mitochondrial Function Variety in the Skin-Derived Fibroblasts of Leber's Hereditary Optic Neuropathy Patients

Shun Yao et al.

Summary: This study found that multiple variations in mitochondrial genes are associated with the heterogeneity, especially phenotypic heterogeneity, in patients with Leber's hereditary optic neuropathy (LHON). Cells with more mtDNA variants showed higher levels of reactive oxygen species (ROS), lower mitochondrial membrane potential, and weaker respiratory function. Multiple mtDNA variants were also associated with different levels of cell viability and apoptosis.

Mitochondrial Dysfunction in Oxidative Stress-Mediated Intervertebral Disc Degeneration

Dian-Kai Wang et al.

Summary: This review discusses the changes in mitochondrial morphology and function during oxidative stress-mediated intervertebral disc degeneration (IVDD), emphasizing the important role of mitochondria in this process. Abnormality in mitochondrial dynamics and dysregulated mitophagy contribute to the accumulation of reactive oxygen species (ROS), leading to cell apoptosis, cell senescence, excessive autophagy, and mitochondrial dysfunction. The excessive ROS production and impaired ROS clearance create a vicious cycle that results in the death of intervertebral disc cells and degradation of the extracellular matrix. Based on the understanding of the role of oxidative stress in IVDD, therapeutic strategies targeting mitochondrial dysfunction are summarized.

Retinal Proteomic Alterations and Combined Transcriptomic-Proteomic Analysis in the Early Stages of Progression of a Mouse Model of X-Linked Retinoschisis

Xiuxiu Jin et al.

Summary: This study used data-independent acquisition-mass spectrometry-based proteomic analysis to identify changes in protein expression related to X-linked retinoschisis (XLRS) in a mouse model. The results showed upregulation of type I interferon-mediated signaling and downregulation of photoreceptor proteins responsible for detection of light stimuli in the early stage (P15). In the early progression stage (P56), positive regulation of Tor signaling was downregulated and the nuclear transcribed mRNA catabolic process nonsense-mediated decay was upregulated. Combined transcriptomic-proteomic analysis revealed functional impairments in detection of visible light, visual perception, and visual phototransduction from P21 to P56.

Pathologically Responsive Mitochondrial Gene Therapy in an Allotopic Expression-Independent Manner Cures Leber's Hereditary Optic Neuropathy

Yi Wang et al.

Summary: Leber's hereditary optic neuropathy (LHON) is a rare inherited blindness caused by mutations in mitochondrial DNA. The traditional chemotherapeutic agent Idebenone only alleviates symptoms, and in situ mitochondrial gene therapy is challenging. A novel pathologically responsive mitochondrial gene delivery vector named TISUH has been developed, showing promising potential for LHON treatment by targeting diseased mitochondria and releasing functional genes for correcting genetic abnormalities.

Summary: The study showed that in cells of sporadic AD patients, there is a decrease in mitochondrial turnover function, lower levels of proteins related to mitochondrial biogenesis and autophagy, higher mitochondrial superoxide levels, and impaired antioxidant defense. These findings suggest a shift in the balance of mitochondrial function in response to cellular stress related to AD development.

Leber's Hereditary Optic Neuropathy Arising From the Synergy Between ND1 3635G>A Mutation and Mitochondrial YARS2 Mutations

Xiaofen Jin et al.

Summary: This study investigated the synergic interaction between LHON-associated ND1 and YARS2 mutations, revealing that the mutations lead to greater mitochondrial defects and increased autophagy. The ND1 mutation altered protein structure and function, while the YARS2 mutation affected protein stability. Cells harboring both mutations exhibited more severe deficiencies in mitochondrial function and showed higher levels of autophagy compared to cells with only one mutation.
