PSME4 Activates mTOR Signaling and Promotes the Malignant Progression of Hepatocellular Carcinoma

Proteasome complexes experience profound structural and functional rearrangements throughout mammalian spermatogenesis

Summary: During spermatogenesis, the proteasome plays a crucial role in regulating cell division processes. The s20S proteasome becomes highly active during meiosis, mainly through 19S activation and PA200 binding. The proteasome population shifts from c20S to s20S during differentiation, and s20S interacts with components of the meiotic synaptonemal complex. In vitro, s20S shows higher trypsin- and chymotrypsin-like activities and displays structural differences between alpha 4 and alpha 4s.

Indirubin-3' -monoxime acts as proteasome inhibitor: Therapeutic application in multiple myeloma

Summary: The study suggests that Indirubin-3'-monoxime (I3MO), a derivative of Indirubin, exhibits anti-tumor activity in multiple myeloma (MM) by sensitizing MM cells to bortezomib-induced apoptosis. I3MO acts as a multifaceted regulator of cell death, inducing DNA damage, cell cycle arrest, and abrogating NF-KB activation. It down-regulates the expression of USP7, promotes NEK2 degradation, and suppresses NF-KB signaling. Moreover, I3MO directly binds to and down-regulates the proteasome activators PA28g (PSME3) and PA200 (PSME4). High expression of PSME3 and PSME4 is associated with poor outcome in relapsed/refractory MM (RRMM). Overall, I3MO represents a promising therapeutic strategy for MM.

Summary: The study revealed that PA200 protein regulates gene expression in neuroblastoma cells by impacting metabolic processes such as glycolysis and mitochondrial function at the transcriptional level. Depletion of PA200 led to metabolic shifts towards glycolysis from oxidative phosphorylation, along with changes in mitochondrial morphology and proteolytic cleavage. These findings suggest a role for PA200 in modulating metabolic changes in response to ATP synthase inhibition in a cellular model.

High PSME1 and PSME2 expression in GC patients is associated with enhanced anti-cancer immunity and potential response to immunotherapy, while PSME3 may act as an oncogene and PSME4 shows promise as a diagnostic indicator for GC.
