Micro-scaffolds as synthetic cell niches: recent advances and challenges

Summary: Micro-fabrication and nano-fabrication provide useful methods for addressing biological questions by mimicking physiological microenvironments. Two-dimensional patterns and three-dimensional scaffolds, obtained through techniques like lithography and direct laser writing, can shape hydrogels, synthetic polymers, and biologically derived materials for (single) cell culture. Micro-scaffolds mimicking cell niches have applications in stem cell renewal, differentiation, and lineage specification, as well as studying single cells in basic cell biology.

Further in vivo studies are needed to explore the potential of these nanomedicines for treating bone sarcomas.

A targeted combinatorial therapy for Ewing's sarcoma

Summary: Ewing's sarcoma is the second most common bone cancer in children and adolescents. Current chemotherapy is often ineffective in relapsed cases, prompting the development of a novel combinatorial therapy with a promising drug candidate named ML111. Polymeric nanoparticles were used to enhance the solubility of ML111, which showed selective activity against EwS cells and synergistic effects with vincristine. In vitro and animal studies demonstrated the superior therapeutic outcomes of ML111-NP in combination with vincristine, suggesting its potential as a new treatment approach for EwS.
