Adipose-Derived Circulating Exosomes Promote Protection of the Pulmonary Endothelial Barrier by Inhibiting EndMT and Oxidative Stress through Down-Regulation of the TGF-β Pathway: A Potential Explanation for the Obesity Paradox in ARDS

Di Qi et al.

Summary: The obesity paradox in ARDS refers to the phenomenon in which obesity is associated with higher morbidity but lower mortality. Adipose-derived circulating exosomal miR-122-5p protects against ARDS by reinforcing pulmonary endothelial barrier through inhibition of EndMT and oxidative stress via down-regulation of the TGF-beta pathway. This provides a potential explanation for the obesity paradox in ARDS and suggests promising prospects for adipose-derived exosomes in cell-free therapies for ARDS.

Exosome-loaded extracellular matrix-mimic hydrogel with anti-inflammatory property Facilitates/promotes growth plate injury repair

Pengfei Guan et al.

Summary: Growth plate cartilage has limited self-repair ability, leading to poor bone bridge formation and limb growth defects in children. The researchers developed an exosome-derived hydrogel that directly supplements extracellular matrix and inhibits inflammation to promote cartilage repair. The hydrogel significantly enhanced extracellular matrix synthesis and facilitated growth plate injury repair through ECM remodeling.

A bacterial extracellular vesicle-based intranasal vaccine against SARS-CoV-2 protects against disease and elicits neutralizing antibodies to wild-type and Delta variants

Linglei Jiang et al.

Summary: This study presents a novel SARS-CoV-2 vaccine candidate based on extracellular vesicles (EVs) that are decorated with the Spike receptor-binding domain (RBD). The vaccine candidate induced high titres of antibodies and demonstrated neutralizing antibody activity against wild-type and Delta variants. Animal experiments showed that hamsters immunized with the RBD-OMV vaccine had better protection against virus infection and experienced less severe pathology.

Human cerebral organoids establish subcortical projections in the mouse brain after transplantation

Xin Dong et al.

Summary: The study demonstrated the successful transplantation of human cerebral organoids into the mouse brain cortex, with the organoids surviving and extending neural projections over 4.5 mm in length within 1 month. The transplanted organoids generated mature neurons and formed synaptic connections with host neurons, integrating into existing neural circuits and impacting the neural behavior of the mice.

Characteristics and Therapeutic Potential of Human Amnion-Derived Stem Cells

Quan-Wen Liu et al.

Summary: Stem cell therapy has the potential to repair tissues and improve functional recovery, but faces challenges such as ethical barriers and immune rejection. Human amnion-derived stem cells (hADSCs) show advantages over other stem cells, including high pluripotency and immunomodulatory effects, making them a promising source for cell therapy and regenerative medicine.

TGFβ signalling is required to maintain pluripotency of human naive pluripotent stem cells

Anna Osnato et al.

Summary: TGF beta signaling is essential for maintaining naive human pluripotent stem cells, with downstream effector proteins SMAD2/3 binding to common sites in both naive and primed cells. Inhibition of TGF beta signaling leads to downregulation of target genes and differentiation of pluripotent cells, with different transcriptional trajectories observed between naive and primed cells. These findings indicate a continuum of TGF beta pathway function in human pluripotency.

Disruption of Tfh:B Cell Interactions Prevents Antibody-Mediated Rejection in a Kidney Transplant Model in Rats: Impact of Calcineurin Inhibitor Dose

Louisa Steines et al.

Summary: In antibody-mediated rejection (ABMR), the formation of donor-specific antibodies (DSA) is correlated with the frequency and expansion of germinal centers (GC) and activated T follicular helper cells (Tfh). High dose cyclosporine A (hiCNI) can suppress the expression of cytokines like IL-21, reducing Tfh activation and preventing the development of DSA and ABMR.
