Semiconducting Polymer Nano-regulators with Cascading Activation for Photodynamic Cancer Immunotherapy

NIR-II Functional Materials for Photoacoustic Theranostics

Summary: Photoacoustic imaging (PAI) is a promising technique for the diagnosis and treatment of diseases. In the second near-infrared (NIR-II) window, PAI can effectively overcome the interference of tissue fluorescence and obtain high resolution images. NIR-II PA contrast agents have been developed to improve the performance of PAI in disease diagnosis and treatment. The applications of NIR-II PA contrast agents in medical imaging, such as cancer imaging and inflammation detection, are discussed.

Activatable Polymeric Nanoprobe for Near-Infrared Fluorescence and Photoacoustic Imaging of T Lymphocytes

Summary: The development of real-time non-invasive imaging probes for assessing infiltration and activation of cytotoxic T cells (CTLs) is essential for predicting the efficacy of cancer immunotherapy. This study introduces an activatable semiconducting polymer nanoprobe (SPNP) for near-infrared fluorescence (NIRF) and photoacoustic (PA) imaging of a CTL activation biomarker, granzyme B. The SPNP can passively target tumors and react with granzyme B in situ to release a dye-labeled peptide, resulting in correlated NIRF and PA signals with the expression level of granzyme B and intratumoral population of CTLs.

Various strategies such as structure-property screening, fluorescence quenching, accelerated heat dissipation, and size-dependent heat dissipation have been discussed to amplify the PA brightness of SPNs for in vivo PA. Additionally, molecular approaches have been introduced to shift the absorption of SPNs for NIR-II PA imaging and PTT in order to improve tissue penetration depth for diagnosis and therapy.
