MIF/SCL3A2 depletion inhibits the proliferation and metastasis of colorectal cancer cells via the AKT/GSK-3 beta pathway and cell iron death

Guan Huang et al.

Summary: This study investigated the mechanisms of migration inhibitory factor (MIF) and solute carrier family 3 member 2 (SLC3A2) in colorectal cancer progression. Results showed that up-regulated expression of MIF and SLC3A2 in colorectal cancer cells promoted cell proliferation, migration, epithelial-mesenchymal transition (EMT), invasion, and inhibited iron death. Co-immunoprecipitation (CoIP) results indicated direct interaction between MIF and SLC3A2. In vivo experiments confirmed that knockdown of both MIF and SLC3A2 inhibited tumor growth and metastasis via the AKT/GSK-3 beta pathway. MIF and SLC3A2 may serve as potential biomarkers for monitoring colorectal cancer treatment.

Ferroptosis: molecular mechanisms and health implications

Daolin Tang et al.

Summary: Cell death can be executed through different subroutines, such as ferroptosis which is an iron-dependent form of non-apoptotic cell death. Ferroptosis can occur through two major pathways, driven by redox imbalance and abnormal expression of redox-active enzymes. The process is precisely regulated at multiple levels, with the transcription factor NFE2L2 playing a central role in anti-ferroptotic defense.
