Functional evidence for biased inhibition of G protein signaling by YM-254890 in human coronary artery endothelial cells

Qianman Peng et al.

Summary: YM-254890 was initially reported as a selective inhibitor for the Gq protein, blocking GDP exchange of GTP on the a subunit of Gq complex. However, further studies revealed that YM-254890 also exhibits inhibitory effects on Gs proteins and biased inhibition on Gi/o signaling, while not affecting non-GPCR-mediated cellular signaling.

Autoantibodies as Endogenous Modulators of GPCR Signaling

Meredith A. Skiba et al.

Summary: Endogenous self-reactive autoantibodies can recognize G-protein-coupled receptors and are often associated with various diseases; they modulate receptor signaling, but the molecular details of their modulatory activity are still unclear. Technological advances have provided new opportunities to understand the function of GPCR AAs, which also helps to explore the potential roles and limitations of therapeutic antibodies in modulating GPCR signaling.

Biased Allosteric Modulators: New Frontiers in GPCR Drug Discovery

Lauren M. Slosky et al.

Summary: GPCRs are important cell surface receptors and drug discovery in this field faces challenges, but these may be overcome by developing biased allosteric modulators. This emerging class of GPCR ligands can modulate receptor signaling and provide opportunities for fine-tuning physiology.

Allosteric modulators enhance agonist efficacy by increasing the residence time of a GPCR in the active state

Anne-Marinette Cao et al.

Summary: The study utilized smFRET to investigate the structural dynamics of metabotropic glutamate receptor mGlu2 and found that positive allosteric modulators or the Gi protein can stabilize mGlu2 in the active state, leading to full receptor activation. This discovery provides important insights into the role of allosteric modulators in mGlu activation.
