Summary: Biological pretreatment is crucial in biorefinery for utilizing lignocellulosic biomass. A yellow laccase-producing strain was studied for its production, purification, and depolymerization and hydrolysis. Yellow laccase production was optimized and partially purified using a single-step aqueous two-phase system. The partially purified laccase showed stability and effectiveness in depolymerizing lignin. Integrated depolymerization and hydrolysis using different laccase loads enhanced sugar yield in all biomass. One-step depolymerization and hydrolysis have significant value in large-scale biorefinery and towards a cleaner ecosystem.

Substrate specificity of a new laccase from Trametes polyzona WRF03

Summary: In this study, various aromatic compounds structurally similar to lignin were tested as substrates for purified laccase from Trametes polyzona WRF03, with ABTS showing the highest specificity. The oxidation of phenolic substrates was found to be influenced by the nature and position of substituent groups on the aromatic nucleus. These findings suggest potential applications of the enzyme in delignification and treatment of wastewater containing dyes and PAHs.

Kinetic characterization of purified laccase from Trametes hirsuta: a study on laccase catalyzed biotransformation of 1,4-dioxane

Summary: The purified laccase from white-rot fungus T. hirsuta L. showed specific activity of 978.34 U/mg and stable activity at pH 4-6 and temperature range of 20-40 degrees Celsius. The enzyme exhibited significant stability for 10 metal ions up to 10 mM concentration, with Cu2+ inducing laccase activity up to 142% higher than the control at 10 mM concentration. The degradation of 1,4-dioxane by purified laccase was accelerated in presence of syringaldehyde, vanillin, ABTS, and guaiacol mediators.
